产朊假丝酵母菌的筛选及其尿酸酶活力分析

摘　　要：用以尿酸作为唯一碳源的YNB培养基初步筛选出两株产朊假丝酵母,并将筛选出的产朊假丝酵母分别用两种培养基对两株菌进行培养,并比较了两种培养基的培养效果,优化了培养条件;从发酵液中分离纯化尿酸酶,并对尿酸酶活力进行测定,改进了测定方法.结果发现,YNB培养基比酵母膏培养基更适合培养所筛选的产朊假丝酵母;两株菌所产尿酸酶有较高活力.Two strains of Candida utilis were screened from the solid medium with uric acid as a sole carbon source. Two kinds of mediums were used to culture Candida utilis. The effects of two mediums on Candida utilis were studied. The culturing condition was optimized. Purified uricase was isolated from the fermentation broth. En- zyme activity of uricase was determined. Determination method of enzyme activity was improved. It was showed that YNB medium was more suitable for culturing Candida utilis than yeast extract medium. Enzyme activity of uricase produced both of the two Candida utilis uricase were high.
