摘  要:目前孔雀石绿类残留是我国水产品质量安全的隐患之一。本研究选用大菱鲆、大黄鱼和南美极白对虾3种阴性样本,利用商品化试剂盒研究水产品中孔雀石绿酶联免疫检测效果,结果表明,样本基质组分的存在会导致检测结果产生显著的偏差,其中蛋白质是主要的干扰因素之一。利用9%三氯乙酸对提取液进行处理,可以简便有效地去除干扰蛋白,显著降低基质干扰程度。在0.5μg/kg的加标浓度下,3种水产样品的平均回收率基本维持在120%~130%,相对标准偏差均小于11.2%,基本达到了实践中筛选检测的要求。Malachite green (MG) is now considered one of the most important hazards in aquatic products. The effi- ciency of enzyme - linked immunosorbent assay (ELISA) for MG in aquatic products was investigated and validated by using Scophthalmus maximus, Pseudosciaena crocea and Penaeus vannamei as samples. The results indicated that, the presence of matrix components could significantly interfere the ELISA, and some proteins were proved to contri - bute greatly to the matrix interference investigated. A pretreatment of the samples with 9 % trichloroacetic acid was demonstrated to be able to remove the proteins effectively, therefore significantly improve the accuracy of the ELISA. At a spiking concentration of 0.5 I^g/kg, the average recovery for the three kind of aquatic products was demonstrated in the range of 120% - 130 % , with all relative standard deviations below 11.2 %, which indicated the application prospect of the technique for fast screening of the MG residue in aquatic products.
