摘 要:目的利用D-半乳糖诱致小鼠学习记忆障碍,探讨荔枝核提取物对学习记忆障碍的改善作用及其机制。方法 50只昆明种小鼠随机分为空白对照组、模型对照组、石杉碱甲组、荔枝核提取物低和高剂量组。上述动物除空白对照组外,均皮下(sc)注射D-半乳糖8周,并于第5周起分别灌胃相应药物。采用水迷宫和自主活动仪测试小鼠学习和记忆能力,测定血清糖基化丙二醛(MDA)的含量、超氨化物岐化酶(SOD)和颅内肿瘤(GSH-PX)活性;结果荔枝核提取物明显改善模型小鼠学习和记忆能力,减少血清糖基化产物(AGEs)含量,同时均可明显提高SOD和GSH-PX活性;降低脑组织MDA、一氧化氮(NO)含量和一氧化氮合酶(NOS)活性,结论荔枝核提取物具有改善D-半乳糖诱致小鼠学习记忆障碍的作用。Objective Using D-galaetose to estabilish the learning and memory obstacles model in mice to investigate the improving function and the machenism of Litchi Seed Aqueous Extracts on this model. Methods D-galactose was injected into mice(50 mice) by hypodermic for 8 weeks to induce learning and memory obstacles, except the control group. Huperzine A and Litchi Seed Aqueous Extracts were administered for 4 weeks by ig at the 5th week of the making model. After huperzine A and Litchi Seed Aqueous Extracts treatment for 4 weeks, using water maze test to determine the ability of mice's spatial learning and memory, the content of nitric oxide ( NO ) and acetylcholine ( Ach ), the activity of nitric oxide synthase (NOS) on the of advanced glycation end products Malondialdehyde (MDA)in serum, Super Oxidase Dimntase (SOD) and Intraeranial tumor(GSH-PX) the activity of in the brain tissue were detected separately. Results Litchi Seed Aqueous Extracts significantly ameliorated the learning and memory obstacles model in mice, decreased the level of in serum, could obviously increase the activ- ity by SOD and GSH-PX in serum and reduced the content of MDA ,NO and activity of NOS in could obviously decrease of serum and activity in brain tissues. Conclusion Litchi Seed Aqueous Extracts possesses improving the [earning and memory effects on the model mice induced by D-galactose,which may be related to inhibiting to much NOS and NO formation and reducing damage on the brain cells.
