蜂蜜对新目安眼用凝胶中更昔洛韦在兔离体角膜中的渗透释药行为的影响

摘　　要：目的研究不同浓度的蜂蜜对新目安眼用凝胶中更昔洛韦在兔离体角膜中的渗透释药行为的影响,优选新目安眼用凝胶中蜂蜜的最佳用量,为新目安眼用凝胶处方设计提供依据。方法以谷胱甘肽缓冲液为释放介质,采用高效液相色谱法测定新目安眼用凝胶中更昔洛韦在兔离体角膜中的累积渗透释药量,绘制累积释药曲线,建立动力学模型,提取累积渗透速率t70%及累积渗透百分率Q8h。结果含5%蜂蜜的新目安眼用凝胶与含7%蜂蜜的新目安眼用凝胶的兔离体角膜渗透速度、兔离体角膜累积渗透百分率差异均无统计学意义,但均优于含3%蜂蜜的新目安眼用凝胶。结论新目安眼用凝胶中蜂蜜的浓度为5%为佳。Objective To study the effects of honey on the rabbits isolated cornea osmotic re- leasing behaviors of Ganciclovir in Mu＇an ophthalmic gel and to optimize the dosage of honey in Mu＇an ophthalmic gel, and then provide the basis for prescription design. Methods The cumulative osmotic medicine releasing amount of Ganciclovir in Mu＇an ophthalmic gel in the isolated cornea of rabbits was determined by HPLC with glutathione buffer as releasing medium. The curves of medicine release were drawn and the kinetics models were established and cumulative osmotic velocity （t70%）and percentage （Q8h） were extracted. Results There were no statistical differences in t70% and Q8h of Ganciclovir between Mu＇an ophthalmic gel containing 5% honey group and Mu＇an Oph- thalmic gel containing 7% honey group, but they were higher than Mu＇an ophthalmic gel containing 3% honey group. Conclusion Mu＇an ophthalmic gel containing 5% honey is optimal in cumulative osmotic velocity（t70%） and percentage（Q8h）.
