Optimization of Yeast Oil Extraction Technique by Response Surface Methodology

摘  要:研究应用响应面法优化黏红酵母油脂提取的工艺。通过Plackett-Burmen试验筛选出对油脂得率有显著影响的因素,再用Box-Behnken试验优化这4个因素的水平。按最优工艺提取,实际油脂质量分数为(50.21±1.39)%,提高了26.19%。用气相色谱-质谱联用仪对得到的油脂进行脂肪酸组成的分析,结果显示该菌株产生的油脂主要由油酸(C18∶1)、软脂酸(C16∶0)、亚油酸(C18∶2)、硬脂酸(C18∶0)和棕榈油酸(C16∶1)组成,其脂肪酸组成与植物油近似,因此可以作为生产生物柴油的原料。The objective of the present study was to apply the response surface methodology to optimize the technique of oil extraction of rhodotorula glutinis. Several factors with significant influences on the yield of oil were screened by Plackett - Burmen experiment, and the degree of their influences were optimized by Box - Behnken experiment. According to the optimal extraction technique, the actual oil mass fraction was(50.21± 1.39) %, increased by 26.19%. The composition of the fatty acid of extracted oil was analyzed by gas chromatography - mass spectrometry (GC -MS), and the results showed that rhodotorula glutinis oil was mainly comprised of oleic acid (C18:1), palmitic acid( C16: O) ,linoleic acid( C18: 2) ,stearic acid( C18: 0) and palmitoleic acid( C16:1 ). The fatty acid composition was similar to that of the vegetable oil, so it could be used as raw materials of biodiesel production.
