摘  要:目的探究小鼠血小板源性生长因子受体样蛋白(PDGFRL)对结肠癌细胞CT-26增殖和迁移的影响。方法调取小鼠PDGFRL基因并表达全长蛋白及两个结构域蛋白。利用磺酰罗丹明B法(SRB)、流式细胞术和损伤-修复实验检测PDGFRL全长蛋白及两个结构域蛋白对小鼠结肠癌CT-26细胞系增殖、迁移、凋亡及细胞周期的影响。结果 PDGFRL全长蛋白及两个结构域对CT-26细胞增殖有明显的抑制作用,促进细胞凋亡,并阻滞细胞周期于G2/M期。尤以结构域Ⅱ效果最为显著。此外,PDGFRL全长蛋白可抑制CT-26细胞的迁移,而两个结构域蛋白对细胞迁移无明显影响。结论 PDGFRL蛋白能有效抑制CT-26细胞增殖,在促进细胞凋亡过程中,结构域Ⅱ可能发挥重要作用。Objective To investigate the effect of platelet-derived growth factor receptor-like protein(PDGFRL) on the growth and migration of colon cancer CT-26 cells. Methods The PDGFRL full-length protein and its two domains were obtained by prokary- otic expression. Sulforhodamine B ( SRB), flow cytometry and wound-healing assays were used to evaluate the effect of PDGFRL and its two domains on the proliferation, apoptosis, migration as well as cell cycle of CT-26. Results PDGFRL and its two domains had the ability to suppress proliferation, promote apoptosis of CT-26, and cause cell cycle arrest at G2/M phase. In addition, PDGFRL in- hibited migration of CT-26, while two domain proteins had no effect on cell migration. Conclution PDGFRL can effectively inhibit CT-26 cells proliferation, and domain Ⅱ protein may play an important role in promoting apoptosis.
