摘 要:目的:通过牙周结扎诱导大鼠牙周炎的发生,观察牙周骨吸收及去除结扎后的自然转归情况。方法:将32只成年雄性SD大鼠随机分为4组(n=8),用结扎丝结扎上颌右侧第一磨牙牙颈部,自身对侧同名牙作为正常对照。A组:结扎7 d;B组:结扎21 d;C组:结扎21 d后去除结扎丝,继续饲养7 d;D组:结扎21 d后去除结扎丝,继续饲养21 d。通过micro-CT扫描与HE染色、抗酒石酸酸性磷酸酶(TRAP)染色观察牙周组织骨量及破骨细胞数目改变特征。结果:A、B组牙龈组织内炎症浸润,破骨细胞生成排列在牙槽骨边缘,牙槽骨高度下降;B组根分叉下骨密度(BMD)降低(P<0.05);C、D组炎症减轻,牙槽骨吸收情况缓解,牙槽骨高度上升。D组与B组相比,BMD升高(P<0.05)。结论:牙周结扎可刺激牙周炎症发生、发展;而去除结扎可阻止炎症发展,促进牙周组织的自行修复。AIM: To observe the bone resorption in rats with ligature - induced periodontitis and the natural turnover after removing the ligation. METHODS : 32 adult male SD rats were randomly divided into 4 groups ( n = 8 ). The right maxillary first molar was assigned to receive wire ligation in the sulcular position, while the contralateral tooth was unligated as the control. The molars in group A were ligated for 7 day; in group B for 21 day; in group C were ligated for 21-days, then the ligation was removed and the rats were fed for another 7 days; in group D were ligated for 21-days, then the ligation was removed and the rats were fed for another 21 days. At the end of the treatment, the animals were sacrificed and the periodontal tissue of the teeth were examined by micro-CT, HE and tartrate resistant acid -phosphatase (TRAP) staining. RESULTS: Inflammatory infiltration in the gingival tissue, osteoclasts lining on the alveolar bone, and decreased alveolar bone height were observed in group A and B. The decrease of bone mineral den- sity (BMD) under the furcation of group B was significant(P 〈0.05). In groups C and D, the inflammation degree were reduced, the number of osteoclasts decreased, and the alveolar bone height increased when compared with those in group B. BMD of group D was significantly higher than that of group B ( P 〈 0.05 ). CONCLUSION : In the ligature-induced periodontitis of rat model, ligation can promote the development of periodontitis, while removal of the ligation can prevent the development of periodontitis, and activate the self-renair of periodontal tissues.
