摘 要:目的:观察脂肪干细胞(adipose-derived sem cells,ASCs)对糖尿病兔种植体周围新骨生成及种植体骨结合的影响,为解决糖尿病缺牙患者的种植修复难题提供新思路。方法:分离培养兔ASCs,并通过多向诱导分化进行鉴定;14只新西兰大白兔成功建立Ⅰ型糖尿病动物模型后,于每只动物的双侧胫骨植入纯钛种植体,并随机在一侧种植体周围植入ASCs与Bio-Oss骨粉的复合物作为实验组,另一侧植入生理盐水与Bio-Oss复合物作为对照。分别于术后第4、8、12周取材,进行Micro-CT扫描、硬组织切片并行丽春红染色,观察植骨区骨质生成情况及种植体骨结合情况。结果:分离培养的新西兰兔ASCs增殖活性高,诱导后可向成脂细胞及成骨细胞分化,将其与Bio-Oss复合并植于种植体周围8周和12周后,成骨效果及种植体骨结合情况均好于对照组,差异有统计学意义(P<0.05)。结论:ASCs能促进糖尿病新西兰兔种植体周围植骨区新生骨生成,并提高种植体的骨结合率。AIM: To investigate the effect of adipose-derived stem cells (ASCs) on bone formation around implant and osseointegration of the implant in diabetic rabbits. METHODS: ASCs were obtained from New Zealand rabbits and identified by multilineage differentiation assays. Diabetic model was established in 14 rabbits by intravenous injection of Alloxan. Pure titanium implants were inserted into the tibia of the rabbits. The composite of ASCs with Bio-Oss was implanted randomly into one tibia around the implant of the rabbits, the eontralateral tibia served as the control. Bone formation was studied by general observation, histological and Micro-CT examination at 4, 8 and 12 weeks after operation, respectively. RESULTS : ASCs showed the ability of self, renewal, proliferation and multi-differentiation. Bone formation and osseointegration were significantly enhanced when ASCs applied at 8 and 12 weeks after implantation (P 〈 0.05 ). CONCLUSION:ASCs can promote bone formation and osseointegration of implant in diabetic rabbits.
