摘 要:目的:探讨衰老大鼠跑台抗阻训练模型的建立。方法:以45只19月龄SD大鼠为实验对象,利用动物跑台,通过大鼠爬坡状态和运动后血乳酸变化确定跑台抗阻训练的爬坡最大角度、跑速及间歇训练时间,在此基础上,以无负重(抗阻训练模型Ⅰ组)、30%(抗阻训练模型Ⅱ组)、50%(抗阻训练模型Ⅲ组)和70%(抗阻训练模型Ⅳ组)最大负重等4种强度进行8周抗阻训练。训练结束后测量骨骼肌形态、横截面及蛋白质含量,评价抗阻训练对衰老大鼠骨骼肌的作用。结果:衰老大鼠跑台抗阻训练方案为跑台适宜坡度为35°,跑速15m/min,每次跑15s,间歇30s,每组4次,组间间歇3min,训练3组为一个循环,每天训练2个循环,隔天训练1次。训练8周后,抗阻训练模型各组肌纤维排列相对整齐,且抗阻训练模型Ⅱ组、Ⅲ组和Ⅳ组实质横截面积和肌蛋白含量均显著大于对照组(P<0.05,P<0.01)。结论:利用动物跑台的动力和坡度结合大鼠负重建立抗阻训练模型可解决大鼠训练的主动性以及对训练强度、次数、组合的定量控制,跑台抗阻训练模型可有效预防和延缓衰老引起的增龄性骨骼肌质量下降。Objective To establish resistance training model for aged rats. Methods 45 19-monthold SD rats were used as the experimental subjects. The maximum climbing angle, speed and the interval of training were determined in accordance to the change in body condition and blood lactic acid of the rats after climbing on an animal treadmill. The rats underwent 8-week resistance training with 0 load (resistance training model Ⅰ),30% maximal load (resistance training model Ⅱ),50% maximal load (resistance training model Ⅲ),and 70% maximal load (resistance training model IV). After 8-week resistance training ,the morphology, cross section area and protein content of the skeletal muscle were observed. Results The optimal resistance training model on treadmill for aged revealed that the muscle fiber arranged neatly,and muscle cross section area (P 〈 0.05)and muscle protein content (P 〈 0.01)increased significantly after S-week resistance training. Each training section included 4-repetition of 15-second 35° climbing on treadmill at the speed of 15 m/min,with the interval of 30 seconds between repetitions,and 3 minutes between sections. Each circle comprised 5 sections,and 2 circles each day were recommended. Conclusion The training model in this study is able to prevent and delay the aging of skeletal muscle effectively.
