摘  要:目的建立BABL/c小鼠感染大肠杆菌O127的模型及TGF-β1的荧光定量PCR方法检测方法。方法通过灌胃的方式建立肠炎性小鼠模型,并利用嵌合荧光定量的方法对其结肠TGF-β1定量分析实验。结果成功建立了肠炎性小鼠模型,荧光定量PCR试验结果表明实验组TGF-β1表达量明显高于对照组。结论利用经口灌胃的方法可以成功的建立肠炎性小鼠动物模型,TGF-β1定量方法可以作为模型建立的评价标准。Objective To establish a mouse model of Escherichia coli O127 infection and to detect the TGF-β1 expression in the colon 0f mouse models by real-time PCR. Methods Thirty-six BALB/c mice ( male: female = 1 : 1 ) were used in this study. Colon inflammation in the mice was induced by intragastric administration of Escherichia coil 0127. The expression of TGF-β1 in the colon tissue was analyzed using fluorescent quantitative PCR. Results The mouse models of colon inflammation were successfully established. The TGF-β1 expression level of the experimental group was sig- nificantly higher than that of the control group. Conclusions Mouse models of colon inflammation can be successfully es- tablished by intragastric gavage of E. coll. Quantitative PCR detection of the TGF-β1 expression can be used as a standard of evaluation of the establishment of the mouse models.
