摘  要:用油菜品种“西南302”(B.napes)为材料,研究了油菜种子贮藏蛋白的分离及部分性质。用 Na2HPO4-NaOH(pH 11.4)缓冲液抽提蛋白。 4%~20%连续梯度SDS—PAGE垂直板电泳表明,该贮藏蛋白组分十分复杂,其分子量范围是 8, 000~ 198,000,带型不仅多,而且含量很不一致,其中低分子量蛋白质(8, 000~ 30, 000)含量占 50%~ 60%以上。凝胶等电聚焦电泳表明,等电点范围为 pH3. 5~ 9,其中酸性蛋白质含量占 65%左右。In this paper, the extraction and characteristics of storsge proteins of 'South west 302' rapeseed(B. napus) were studied systematically using Na2HPO4-NaOH buffer (pH 11.4) to extract proteins, three key problems to study plant seed proteins were solved. The results of 4% - 20% gradient PAGE on vertical plate showed that the compositions of rapeseed storage protein were complicate. Molecular weight (MW) of the proteins were between 8.000~198,000. There were 19 bands on the SDS-PAGE and their contents were different significantly. The contents of lower MW proteins of 8,000-30,000 were up to 60%, Gel isoelectrofocusing electrophoresis (IEF) suggested that the pI of these proteins was between pH 3.5-9.0. Among them, acidic proteins were 65% of total protein.
