摘  要:目的研究大鼠闭合性脑损伤后基质金属蛋白酶9(MMP-9)、缝隙连接蛋白43(Connexin-43)、血管内皮生长因子(VEGF)的表达及其不同损伤时间的相关性。方法清洁级成年雄性SD大鼠60只,随机分为正常对照组(n=5),假损伤组(n=5)和损伤组(损伤后1、6、12h及1、2、3、5、7、10、14d共10组)。采用免疫组织化学染色法(SP法)检测MMP-9、Connexin-43在闭合性颅脑损伤后不同时间的表达情况;采用酶联免疫吸附法(ELISA法)检测外周血中VEGF的含量。结果脑损伤6h后MMP-9表达明显增高,于24h达高峰,维持较高水平至7d;脑损伤后Connexin-43表达逐渐增加,伤后6h即可见表达增加,3d达高峰,14d基本正常;损伤组VEGF在损伤后6h明显增高,3dVEGF表达水平升至高水平,并维持高水平至10d。结论大鼠闭合性颅脑损伤后MMP-9、Connexin-43、VEGF表达与脑损伤时间密切相关,且各自呈一定的时序性变化规律,可望为法医学颅脑损伤时间的推断提供参考。Objective To investigate the expression changes of matrix metalloproteinase-9(MMP-9),Connexin-43,vascular endothelial growth factor(VEGF) in rats with closed traumatic brain injury.Methods Sixty clean animal SD rats were randomly divided into control group(n=5),sham group(n=5) and the injury groups(the injury 1,6,12 h and 1,2,3,5,7,10,14 d of 12 groups).The expression of MMP-9,Connexin-43 were assessed through the immunohistochemisty SP method.The plasma levels of VEGF were examined using ELISA.Results The expression of MMP-9 progressively increased at 6 h,reach the maximum at 24 h and kept at high level up to 7 d after TBI.Connexin-43 expression progressively increased,enhanced at 6 h,peaked at 3 d and normalized in 14 d.The expression of VEGF in peripheral blood progressively increased at 6 h,reached a higher level at 3 d and kept at high level up to 10 d.Conclusion The results showed a close relationship between the expression of VEGF,MMP-9,Connexin-43 and the progress of brain injury.The time-dependent expression feature might provide evidence for brain injury in forensic detection.
