摘 要:以小鼠为实验模型,通过对经X射线照射后 12~32d的小鼠染色体G带核型分析,发现与白血病发生有密切关系的特异性染色体畸变是15号染色体三体和 12号染色体E-F区的易位。在细胞遗传学分析基础上,用 PCR-SSCP方法检测了与特异性染色体畸变有关的受照射小鼠c-Inyc原癌基因和p(53)抑癌基因的结构变化。选用c-myc基因PCR-SSCP试剂盒对正常和受照射小鼠进行了分析,实验结果未发现特异性改变。用 PCR-SSCP和 Southern Blot方法又分析了部分胸腺瘤小鼠的p(53)基因变化,结果发现,个别样品中在第5、7、8对外显子上有点突变发生,但未发现有相同的 p(53)基因改变.It was detected by chromosome G-band analysis of experimental mice after 12-32 days of x-ray irradiation that the characteristic chromosome aberrations of trisomy 15 and translocation in E--F regions of chromosome 12 were related closely to leukemia generation. Based on the cytogenetic analysis, tile structural changes of c-myc protooncogene and p53 anti-oncogene were examined by PCR-SSCP and Southern Blot methods. The significant mutations have hot been found by the examination of c-myc gene in thymocytes of normal and irradiated mice. The analysis resuts of p53 gene in several thymoma mice showed that there were not common p53 gene mutations, except a few samples with point mutations on the exon 5, 7 or 8.
