不同激素配比对明党参叶片愈伤组织诱导的影响及其总香豆素的含量测定

摘　　要：目的:考察不同激素配比对明党参叶片愈伤组织诱导的影响;建立测定明党参叶片愈伤组织中总香豆素含量的方法。方法:采用L9(34)正交试验设计不同激素对明党参叶片愈伤组织的诱导体系;以出愈率和生长状况为指标,考察不同激素配比对愈伤组织诱导的影响。以珊瑚菜内酯为指标性成分,检测波长为268nm,采用紫外-可见分光光度法测定愈伤组织中总香豆素的含量。结果:以MS培养基+2,4-二氯苯氧乙酸0.5mg/L+6-糠氨基嘌呤0.2mg/L+1-萘乙酸1.5mg/L+6-苄氨基腺嘌呤0.5mg/L的激素组合对明党参叶片愈伤组织的诱导效果较好,培养14d左右的出愈率即达到100%,且生长状况也较好。珊瑚菜内酯的质量浓度在4.06～11.60μg/L范围内与吸光度呈良好的线性关系(r=0.9948);平均加样回收率为103.15%,RSD=2.04%(n=6)。结论:该方法操作简便、快捷,结果准确,可用于明党参叶片愈伤组织中总香豆素的含量测定,也可为明党参愈伤组织诱导的激素配比选择提供依据。OBJECTIVE： To investigate the effects of different proportions of hormone on leaf callus induction of Changium smyrnioides, and to establish a method for the determination of the total coumarins in leaf callus induction of C. smyrnioides. METHODS：The system of different hormones inducing leaf callus of C. smyrnioides was designed with L9 （34） orthogonal test; with callus induction rate and growth as index, the effects of different proportions of hormone on leaf callus induction. With phellopterin as index component, the content of coumarins in callus was detected by UV spectrophotometry at 268 nm. RESULTS： MS culture medium＋2,4-D 0.5 mg/L＋KT 0.2 mg/L＋NAA 1.5 mg/L＋6-BA 0.5 mg/L showed sound induction effect on leaf callus of C. smyrnioides; callus induction rate reached 100% about 14 days later, and leaf callus of C. smyrnioides had grown well. The linear range of phellopterin was 4.06-11.60 μg/L（r=0.994 8） with an average recovery of 103.15% （RSD=2.04% ,n=6）. CONCLU- SIONS ： The method is simple, rapid and accurate for the content determination of the total coumarins in leaf callus of C. smyrnioides. It also can provide reference for the selection of hormone proportion for leaf callus induction of C. smyrnioides.
