摘  要:目的:通过比较食蟹猴与人淋巴结中不同类型免疫细胞的差异,为临床前药物安全性研究由动物数据外推至人提供科学支持。方法:收集2004-2011年共31项食蟹猴毒性研究试验中阴性对照组104只食蟹猴以及河北医科大学提供的11例人的正常淋巴结,针对B细胞(CD2+0)、T细胞(CD3+,CD4+,CD8+)以及巨噬细胞(CD6+8)进行免疫组织化学检测。结果:食蟹猴及人淋巴结中CD20和CD68为强阳性表达。与人淋巴结不同,食蟹猴淋巴结CD3为阳性或中度阳性,CD4为阳性或弱阳性表达,而CD8为阴性表达。结论:食蟹猴B淋巴细胞与巨噬细胞在人和猴之间非常类似,显示出一定的遗传相似性,但食蟹猴T细胞与人之间存在较大差异,尤其是细胞毒性T细胞。Objective: To provide scientific support for transferring animal data of preclinical safety study to human, we compared the differences in different types of immune ceils of lymph node between eynomolgus monkeys and human. Methods: The normal lymph nodes of cynomolgus monkeys (n = 104) and human (n = 11 ) were collected from animals in control groups of 31 toxicity studies and Hebei Medical School, respectively. Immunohis- tochemistry was conducted to detect B cells ( CD20^+ ), T cells ( CD3^+ , CD4^+ , CD8^+ ) and macrophages ( CD68^+ ). Results : There was a strong positive stain of lymph nodes for CD20 and CD68 in cynomolgus monkey and human lymph nodes. In comparison with human, there was weak stain for CD3 and CD4 , and negative stain for CD8 in cynomolgus monkeys. Conclusion: B lymphocytes and macrophages in human and monkey are very similar, showing a high degree of genetic similarity. However, there is a huge difference in T lymphocytes between the eynomolgus monkeys and human, especially eytotoxic T-cells.
