白鲜皮化学对照品黄柏酮的提取工艺研究

摘  要:目的优选出白鲜皮中对照品黄柏酮的最佳提取工艺。方法采用高效液相色谱法测定黄柏酮含量,采用单因素水平考察乙醇溶剂浓度(65%~95%)、料液比(1∶6~1∶12)、提取时间(1~4 h)、提取次数(1~4次)对黄柏酮提取率的影响;分别对乙醇浓度、料液比、提取时间、提取次数等因素,设计四因素、三水平正交试验法,确定黄柏酮最佳的提取工艺。结果在单因素考察中,乙醇的最佳浓度范围是75%~95%,最佳的料液比范围是1∶8~1∶12,最佳的提取时间范围是1~3 h,最佳的提取次数范围是1~3次。正交试验极差结果分析表明,所考察的因素中,对白鲜皮中黄柏酮提取率的影响程度,从大到小依次为:料液比>提取次数>溶剂浓度>提取时间;黄柏酮的最佳提取工艺为用8倍量95%的乙醇提取2次,每次1 h。按照优化后的提取工艺,黄柏酮的提取率为4.192 mg/g。结论本提取工艺可行,具有工艺流程简单、提取率高的优点。Objective To optimize the best extraction technique of obacunone, a chemical reference substance in Baixianpi (Cortex Dictamni). Methods The content of obacunone was determined by applying HPLC. The influences of ethanol concentration (65% to 95% ), solid-liquid ratio ( 1:6 to 1 : 12), exaction time (1 h to 4 h) and extraction times (1 time to 4 times) on the extraction ratio of obacunone were observed by using single factor test. A four-factor, three-level orthogonal experiment method was designed respectively according to factors of ethanol concentration, solid-liquid ratio, exaction time and extraction times for determining the best extraction technique of obacunone. Results In the single factor test, the best ethanol concentration was from 75 % to 95 %, the best range of solid-liquid ratio was from 1:8 to 1:12, the best range of exaction time was from 1 hour to 3 hours, and the best range of extraction times was from 1 time to 3 times. The range analysis of orthogonal experiment showed that all influence factors were, in a descending order, solid-liquid ratio, extraction times, ethanol concentration and exaction time according to their influence degree. The best extraction technique of obacunone was as follows: 8 times amount of 95% ethanol was used to extract 2 times and each time for one hour in the extraction. The extraction ratio of obacunone was 4. 192 mg/g by this optimized extraction technique. Conclusion This extraction technique is feasible and simple with higher extraction ratio.
