激光氧液对癫痫持续状态大鼠海马神经元巢蛋白表达的影响

摘  要:目的观察激光氧液对血氧分压及戊四氮(PTZ)所致癫痫持续状态(Status epilepticus,SE)大鼠海马神经元巢蛋白(nestin)表达的影响。方法 28日龄雄性SD大鼠抽取腹主动脉血送检血氧分压。28日龄雄性SD大鼠以PTZ建立SE模型,随机分为PTZ+激光氧液组、PTZ+葡萄糖(GS)组和生理盐水(NS)+激光氧液组、NS+GS组,分别给予激光氧液和5%葡萄糖液尾静脉注射,于模型制作成功后第0天、3天、7天、10天、14天通过免疫组织化学方法观察海马区Nestin阳性细胞数。结果注射激光氧液后0.5小时、1小时、1.5小时、2小时各时间点血氧分压与注射激光氧液前比较均明显升高,分别为:(102.25±8.87)mmHg、(144.00±7.03)mmHg、(112.25±8.86)mmHg vs(96.88±7.12)mmHg(P<0.05),注射后1小时达到最高。PTZ+GS组大鼠nestin阳性细胞数在SE后3天、7天、10天、14天较NS+GS组明显增高,分别为(10.83±2.07)个/视野、(13.33±2.16)个/视野、(17.17±1.72)个/视野vs(18.50±2.17)个/视野(P<0.05)。PTZ+激光氧液组大鼠nestin阳性细胞数在7天、10天、14天比PTZ+GS组增多,分别为(13.33±2.16)个/视野、(17.17±1.72)个/视野vs(18.50±2.17)个/视野(P<0.05)。结论激光氧液能提高大鼠血氧分压;激光氧液可以增强SE后大鼠海马神经元nestin的表达,促进神经元再生。Objective To investigate the therapeutic mechanism of laser oxygen liquid to status epilepticus(SE) by observing the influences of laser oxygen liquid to the blood oxygen partial pressure of normal rats and the expression of neuroepithelial stem protein (nestin) of hippocampal neurons of rats following pentylenetetrazolqnduced SE. Methods The blood oxygen partial pressure of 28-day-old Sprague Dawley(SD) healthy male rats were checked by extraction of abdominal aortic blood postnatal day 28 (P28) SD rats received pentetrazole(PTZ) intra-abdominaly to make the model of SE. The rats were divided randomly into PTZ + laser oxygenate fluid group, PTZ + glucose solution(GS) group,NS+laser oxygenate fluid group and normal saline(NS)+ GS group, the number of each group was 40 rats. Each group was injected with laser oxygen liquid and glucose of 50% from tail vein. After the rat models were success{ully established for 0 day,3 days, 7 days, 10 days and 14 days, the expressions of nestin in hippocampal were observed by immunohistochemical method. Results The blood oxygen partial pressure showed obvious increase after the laser oxygen liquid was injected for 0.5 h,1 h,1.5 h,2 h in comparing to that without the injection,respectively (102.25±8.87) mmHg,(144.00±7.03) mmHg, (112. 25± 8. 86) mmHg,(96.88±7.12) mmHg(P〈0.05). The blood oxygen partial pressure was the highest after the laser oxygen liquid was injected for 1 hour. The number of nestin- positive ceils in the rats of PTZ+GS group with SE after 3 d,7 d,10 d,14 d increased obviously comparing to that of NS±GS group,respectively (10.83±2.07) /view, (13.33±2.16) /view,(17.17±1.72) /view,(18.50±2.17) /view ( P G0.05). The number of nestin-positive ceils in the rats of PTZ laser oxygen liquid group after 7 d, 10 d, 14 d increased comparing to that of PTZ+ GS group, respectively ( 13.33 ± 2.16) /view, ( 17.17 ± 1.72) /view, ( 18.50± 2.17) /view( P G0.05). Conclusion
