摘 要:目的建立肝癌的动物移植瘤模型。方法将BALB/c小鼠随机分为模型对照组和肝组织注射组。先在小鼠腹腔内传代培养肝癌细胞株H22,抽取小鼠腹水,提取肝癌细胞悬液,不同时间段多次接种于小鼠腹腔中。颈椎脱臼处死存活的小鼠,取肝组织,计算肝指数,苏木精-伊红(HE)染色后观察肝组织病理变化。结果末次接种后30 d肝组织癌变率为67.9%,45 d肝组织癌变率为80.8%。结论直接接种小鼠肝组织可以建立肝癌移植瘤动物模型,但该方法有待进一步改良。Objective To establish the animal transplanted tumor model of hepatocellular carcinoma. Methods BALB/c mice were randomly divided into the model control group and the liver tissue injection group. Firstly, hepatocellular carcinoma cell line H22 was subcuhured in mice peritoneum ,extracting ascites and liver cell suspension ,inoculating repeatedly mice in different time periods. Then, the survival mice were killed by cervical dislocation for extracting liver tissue, calculating the liver index and observing the pathological changes of liver tissue after HE staining. Results The carcinogenesis rate in mice liver tissues on 30,45 d after last inoculation were 67.9% and 80.8% respectively. Conclusion Liver cancer animal model can he established by directly inoculating mice liver tissue ,but the method needs to be further improved.
