摘　　要：目的探讨miR-25在胃癌组织中的表达及与临床病理因素和预后之间的关系。方法用茎-环RT-qPCR检测86例胃癌组织和20例正常胃组织中miR-25的表达情况,分析miR-25的表达与胃癌临床病理特征和预后的关系。结果茎-环RT-qPCR能特异扩增miR-25。miR-25在胃癌组织中的表达量高于正常胃组织(7.495±1.073vs 1.083±0.126,P<0.001);在有淋巴结转移的胃癌组织中的表达高于无淋巴结转移的胃组织(8.589±0.964 vs 5.649±1.257,P<0.001);在TNM分期Ⅲ～Ⅳ期胃癌组织中的表达高于Ⅰ～Ⅱ期(8.117±1.002 vs 6.273±1.215,P<0.001)。miR-25高表达组的中位生存期和3年生存率低于miR-25低表达组(34.9个月vs 43.8个月,38.7%vs66.2%,P=0.006)。结论胃癌组织高表达miR-25,miR-25可能是胃癌的诊断、预后判断标志和治疗靶标。Objective To explore the expression of miR-25 in gastric cancer and study its relationship with elinicopath- ologie features and prognosis of gastric cancer. Methods Expression of miR-25 in 86 gastric cancer tissues and 20 normal con- trols were examined by stem-loop RT-qPCR. The relationship between miR-25 expression and clinicopathologieal characteristics and prognosis in gastric cancer were analyzed. Results miR-25 was specifically amplified by stem-loop RT-qPCR. The expres- sion of miR-25 in gastric cancer tissues was higher than that in normal gastric tissues（7. 495 ± 1. 073 vs 1. 0834 ±0. 126 ,P 〈 0. 001 ） ; it was significantly higher in gastric cancer tissues with lymph node metastases than those without lymph node metasta- ses （ 8. 589±0. 964 vs 5. 649± 1. 257,P 〈 0. 001 ） ; and it was significantly higher in cases of TNM stage Ill - IV than in those of TNM stage Ⅰ - Ⅱ （ 8.117 ±1. 002 vs 6. 273 ± 1. 215 ,P 〈 0. 001 ）. The median survival period and 3-year accumulative sur- vival rate in high miR-25 expression group was significantly lower than that in low miR-25 expression group （34.9 mons vs 43.8 mons,38.7% vs 66.2 % ,P = 0. 006）. Conclusion miR-25 is highly expressed in gastric cancer tissues. It might be a marker for the diagnosis and prognosis judgment of gastric cancer and a promising target for therapy.
