摘  要:目的探讨灯盏细辛注射液对大鼠脑缺血/再灌注损伤的神经保护作用。方法成年雄性Wistar大鼠24只,应用线栓法经左侧颈外-颈内动脉插线建立缺血2h再灌注48h模型,灯盏细辛注射液腹腔注射(3.6mg/kg)干预治疗,苏木精-伊红(HE)染色观察海马神经元形态,TUNEL法检测神经元凋亡,免疫组化染色法检测IGF-1表达。结果经灯盏细辛注射液干预后,胰岛素样生长因子-1(IGF-1)蛋白表达明显升高,海马神经元显著增加,细胞凋亡明显减少,大鼠认知功能明显改善。结论灯盏细辛注射液可通过上调IGF-1的表达,有效地抑制细胞凋亡,改善学习记忆能力。Objective To study the neuroprotective effect of fleabane injection on cerebral ischemic reperfusion injury in rats. Methods Total of 24 adult healthy male Wistar rats were used to established cerebral ischemia 2h and reperfusion 48h expression models by inserting a thread intraluminally from the left external-internal carotid artery, and then treated by injecting fleabane injection intraperitoneally (3.6 mg/kg). The morphology of neuron was observed by HE stain and the neuronal apoptosis was detected by TUNEL assay, while the expression of insulin-like growth factor-1(IGF-1) in hippocampus were determined by immunohistochemical technique. Results After treated with fleabane injection, the expression of IGF-1 and the number of neurons in hippocampus increased with the cognitive ability improving significantly. Conclusion Fleabane injection might improve the memory ability by inhibiting neuronal apoptosis and up-regulating the expression of IGF-1.
