摘  要:目的观察生长期小鼠锌缺乏条件下肾脏细胞凋亡相关因子BCL-2表达的变化,进而探讨锌离子对肾细胞凋亡的影响。方法根据小鼠精神状况、摄食量、体质量增长及血清锌含量变化确定模型构建是否成功。用免疫荧光和蛋白印迹技术检测肾组织中BCL-2的表达。结果缺锌小鼠出现典型缺锌症状,体质量下降,摄食量减少,血清锌含量明显降低(P<0.05)。在肾脏BCL-2定位表达于肾小管和集合管上皮细胞胞质内,锌缺乏小鼠肾脏BCL-2的表达量明显低于对照组(P<0.05)。结论生长期锌缺乏可使肾组织中BCL-2蛋白表达下调,有可能促进肾细胞凋亡的发生。Objective To observe the expression of BCL-2 in kidney of the mouse with zinc deficiency and to inves- tigate the effect of zinc on the apoptosis of kidney. Methods To evaluate activity, diet intakes, weight and plasma zinc concentration of mice in order to determine whether the animal model function. Immunofluorescence and West- ern blot were applied to observe and to measure the expression of BCL-2 in the kidney of the mouse. Results Zinc deficient mouse displayed typical symptoms of zinc depletion, body weight, and food intake were obviously re- duced. Plasma zinc concentration of the mouse was obviously decreased. Immunofluorescence staining showed that BCL-2 was expressed in the intracytoplasm of epithelial cell of renal tubules and collecting ducts. Western blot showed as compared with control groups, the expression of BCL-2 was obviously decreased. Conclusions Zinc de- ficiency down-regulates expression of BCL-2 protein and promotes apoptosis of renal cells.
