CyclinD1在皮肤病理性瘢痕上皮和瘢痕癌中的表达及意义

摘　　要：目的探讨CyclinD1及其mRNA在皮肤病理性瘢痕及瘢痕癌中的表达及意义。方法激光扫描共聚焦显微技术,对病理性瘢痕和瘢痕癌组织进行CyclinD1免疫荧光双标记。采用免疫组织化学(SP)法分别检测正常皮肤组织、病理性瘢痕和瘢痕癌三组组织中CyclinD1蛋白的表达。采用核酸分子原位杂交技术检测三组组织中CyclinD1 mRNA的表达。结合图像分析,分别计算被检组织中所检各项指标的平均光密度和阳性面积,所有数据运用SPSS16.0软件包进行统计学分析。结果 CyclinD1在病理性瘢痕上皮中呈现较弱荧光,为弱阳性表达;在瘢痕癌组织中呈现较强荧光,为强阳性表达。CyclinD1蛋白、CyclinD1 mRNA在正常皮肤表皮呈阴性表达,在病理性瘢痕上皮中呈弱阳性表达,在瘢痕癌组织中呈强阳性表达。瘢痕癌组的表达(即阳性面积和平均光密度)与正常皮肤组及病理性瘢痕组相比较,差异均有统计学意义(P均<0.01);但正常皮肤组与病理性瘢痕组相比较,差异无统计学意义(P>0.05)。在瘢痕癌组织中,CyclinD1与CyclinD1 mRNA的表达呈正相关。结论 CyclinD1蛋白及其mRNA在病理性瘢痕上皮中的表达无特殊意义,其与瘢痕癌的发生有相关性。Objective To detect the expressions and analyze the significance of CyclinDl and CyclinDlmRNA in skin scar and skin scar carcinima. Methods CyclinD1 in the pathologic scar and carcinoma of scar stained by immunofluorescence dual staining was observed under laser confocal microscopy. Immunohistochemistry （IHC） for CyclinD1 were used in normal skin, pathologic scar and carcinoma of scar, respectively. The ex-pression of CyclinD1 mRNA were detected by in situ hybridization. The optical density and positive area indi-cators seized in the inspection organization were calcutated combing with the analysis of image, and all data was input into the computer and statistically analyzed with SPSS （16.0） software. Results The dual labe-ling of immunofluorecence of CyclinD1 showed weak positive in pathologic scar, while in the carcinoma of scar were strong positive. IHC and ISH for mRNA of CyclinD1 showed negative or weak positive in the normal epidermis and pathological scar, but strong positive in the carcinoma of scar. The expression coverage and intensity between the carcinoma group and normal skin group or pathologic scar group were statistical differ- ence （P 〈0.01 ） ; but without statistical difference between normal skin group and pathologic scar group （P 〉0.05）. Correlated analysis showed： It was positive correlation the expression levels of CyclinD1 and its mRNA in the carcinoma of scar. Conclusion The expression of CyclinD1 protein and its mRNA in the pathological scar epithelium not shown its special significance, but may be related to the occurrence of scar carcinoma.
