提高淡水发光菌检测急性毒性灵敏度研究

摘  要:分别以不同配方的淡水发光细菌青海弧菌Q67冻干粉A、B作为测试菌剂,依据冻干粉菌液与农药及重金属化合物作用15min的半效应浓度(Ecso)的大小,考察4℃冷藏菌液和减少菌液量对冻干粉菌液检测灵敏度的影响。结果表明:采用4℃冷藏菌液、减少菌液量2种方法都可提高冻干粉菌液对农药和重金属化合物的检测灵敏度,提高量为10%~80%;冻干粉菌液检测农药的最佳冷藏时间为24.48h,检测重金属化合物的最佳冷藏时间为48、72h。Two different formulated freeze-dried powders A and B of freshwater luminescent bacteria V. qinghaiensis Q67 were used as test microbe agents. Half maximal effective concentration (ECho) values of the reaction between freeze-dried powder prepared bacterial solutions and pesticides or heavy metal compounds at 15 minutes were adopted to assess the effects of 4 ~C refrigeration and volume reduction of the bacterial solutions on test sensitivities of V. qinghaiensis Q67. The results showed that 4℃ cold storage and reducing volume of bacterial solutions could improve the sensitivities of V. qinghaiensis Q67 to pesticides or heavy metal compounds by 10% to 80%. The best cold storage durations of the bacterial solutions were found to be 24 and 48 hours for monitoring pesticides, whilst 48 and 72 hours for detecting heavy metal compounds, respectively.
