摘 要:目的探讨依达拉奉(edaravone)对小鼠视网膜急性光损伤(retinal photodamage,RP)的保护作用。方法将42只成年雄性BALB/c小鼠随机分为正常对照组(C组,n=6)和实验组(n=36),后者再分为光损伤组(L组)、edaravone组(E组)、生理盐水组(NS组),其中E组和NS组是同一只动物的不同眼别,即右眼注射edaravone,左眼注射生理盐水。实验组采用(8 000±300)lux白色荧光连续照射4h建立小鼠RP模型,于光照后暗修复0、24、72h时取材,视网膜组织切片HE染色观察视网膜形态及外核层(outer nuclear layer,ONL)厚度,免疫组织化学法染色检测原癌基因c-Fos、半胱天冬酶-1(Caspase-1)蛋白的表达变化,以此评价edaravone对RP的保护作用。结果与对照组相比,实验组视网膜ONL厚度均随光照后暗修复时间延长而变薄,E组与L组、NS组同一时间点比较,视网膜ONL变薄程度明显减轻(P<0.05)。实验组于暗修复0h即可在视网膜ONL检测到c-Fos、Caspase-1蛋白表达,二者表达高峰分别位于暗修复0、24h(P<0.05);同一时间点,E组视网膜c-Fos、Caspase-1蛋白表达较L组和NS组均明显降低(P<0.05)。结论 RP可引起视网膜ONL变薄,c-Fos、Caspase-1蛋白表达增加,edaravone通过抑制二者表达间接对RP起到保护作用。Objective To explore the protective effect of edaravone on acute retinal photodamage (RP) in mice. Methods Totally 42 adult male BALB/c mice were randomly divided into normal group (n = 6) and experimental group ( n = 36). The latter was subdivided into light group (group L, n = 18), edaravone group (group E, n 18), and saline group (group NS, n=18). Mice in experimental group were exposed to (8 000-300) lux white fluorescent continuously for 4 hours to establish retinal RP model. Mice were killed at 0, 24, 72 h of dark repair after light exposure. HE staining of retinal tissue sections was used to observe morphologie changes of the retina and to measure thickness of retinal outer nuclear layer (ONL). Immunohistochemieal staining was used to test c-Fos and Caspases-1 expressions. All results were combined to evaluate the protective effect of edaravone on RP. Results ONL became thinner in experimental group with longer dark repair after light exposure than that in normal group. At the same time point, compared to group L and group NS, the thickness of ONL was significantly preserved in group E (P^0.05). At 0 h dark repair, expressions of c-Fos and Caspase-1 protein could be detected in ONL in each experimental group. Each peak of them was at 0 h and 24 h dark repair, respectively. At the same time point, expressions of both c-Fos and Caspase-1 protein were decreased significantly in group E compared to group L and group NS (P-~0.05). Conclusion RP can decrease the thickness of ONL and upregulate the expressions of c-Fos and Caspase-1 protein. Edaravone can protect indirectly the retina from RP by inhibiting c-Fos and Caspase-1 expressions.
