摘  要:目的筛查RET融合基因在鼻咽癌组织和细胞系中的表达情况。方法利用组织芯片通过荧光原位杂交(FISH)和反转录聚合酶链式反应(RT-PCR)技术检测137例鼻咽癌组织和6株常见鼻咽癌细胞系中RET融合基因的表达情况。结果在鼻咽癌组织和细胞系中均未发现RET基因出现断裂融合,同时在细胞系中亦未发现KIF5B/RET基因的表达。结论鼻咽癌中未发现RET融合基因的表达,RET基因在鼻咽癌发生、发展中并未起主导作用,或许不能作为治疗鼻咽癌的靶点,因此需要进行鼻咽癌全基因组分析。Objective To screen the expression of RET fusion gene in nasopharyngeal carcinoma specimens and cell lines. Methods RET fusion gene in tissue microarray and cell lines were detected by fluorescence in situ hybridization (FISH) and reverse transcription polymerase chain reaction (RT-PCR). Results One hundred and thirty-seven cases of nasopharyngeal carcinoma tissues and 6 strains of common nasopharyngeal carcinoma cell lines were screened by fluorescence in situ hybridization. However, no RET fu- sion gene were found in nasopharyngeal carcinoma specimens and cell lines, and further RT-PCR examination of KIF5B/RET fusion gene proved the same. Conclusion There is no RET fusion gene in nasopharyngeal carcinoma, suggesting RET oncogene does not play a leading role in the development of nasopharyngeal carcinoma. RET oncogene may be not a suitable target in the treatment of nasopharyngeal carcinoma, which prompts the existence of other molecular mechanism in the occurrence and development of nasopharyngeal carcinoma. A genome-wide analysis of nasopharyngeal carcinoma is quite essential for understanding the unique biological features of nasopharyngeal carcinoma.
