摘  要:目的观察CD151在大鼠后肢缺血模型中的表达以及对Rac1和Cdc42信号通路的影响,进一步研究CD151促血管新生的分子机制。方法将实验大鼠随机分为4组,分别给予生理盐水,rAAV-GFP和rAAV-CD151分点注射至大鼠左后肢肌肉组织内,基因转染1 w后行股动脉切除术建立大鼠后肢缺血模型,假手术组不切除股动脉。6 w后用Western印迹方法检测各组大鼠缺血肢体局部CD151以及Rac1和Cdc42的表达。结果术后5 w,Western印迹检测CD151组的CD151蛋白表达水平明显高于其余各组(P<0.05),同时CD151组p-Rac1/Cdc42蛋白表达水平明显高于其余各组(P<0.05)。结论 CD151蛋白表达可以激活Rac1和Cdc42信号通路,促进缺血组织的血管新生。Objective To investigate the effects of CD151 mutant gene delivery on Cdc42 and Racl in the model of rat hind-limb ischemia. Methods Rats were randomly divided into 4 groups. Rats in 3 groups were injected with saline, rAAV-GFP, or rAAV-CDISI respectively ( n = 6/group). After 1 week of transfection,the femoral arteries of the rats were excised to establish the model of left hind-limb ischemia. Sham-operated animals underwent the same surgical prncedure without femoral artery resection. The expression of CD151 and phos- phor-Racl/Cdc42 in the ischemic tissue was detected by Western blot. Results The expression of CD151 protein was increased significantly in rAAV-CD151 groups compared with other groups (P 〈 0. 01 ). The expression of phosphor-Racl/Cdc42 protein were increased significantly in rAAV-CD151 groups compared with other groups (P 〈0. 01 ). Conclusions CD151 gene transfer can activate Racl and Cdc42 in rat ischemia hind-limb.
