摘 要:目的研究壳聚糖磷酯酰胆碱对大鼠海马内注射Aβ25-35诱导的阿尔茨海默病(AD)诱导型一氧化氮合酶(iNOS)、IκB激酶β(IKKβ)表达和AD大鼠学习记忆能力的影响。方法双侧海马注射Aβ25-35建立AD炎性模型,采用Morris水迷宫和免疫组化方法分别观察大鼠的学习记忆能力和炎性因子iNOS、IKKβ表达情况。结果模型组大鼠的平均逃避潜伏期(AEL)、单位时间内跨越原平台次数和在原平台象限时间与对照组和药物组比减少,差异有统计学意义(P<0.05)。模型组大鼠海马iNOS和IKKβ阳性细胞表达明显高于对照组(P<0.01),药物组iNOS和IKKβ阳性细胞表达较模型组显著下降(P<0.05)。结论壳聚糖磷酯酰胆碱对AD大鼠脑海马部位炎症反应有一定的抑制作用,及提高AD大鼠学习记忆能力的作用。Objective To determine the effect of Chitosan-Phosphatidylcholine on the expression of inducible nitric oxide synthase (iNOS) , IKB kinaseβ( IKKβ) in hippocampus and the learning and memory ability of Alzheimer's disease(AD)rat induced by Aβ25-35. Methods The Aβ25 -35 was injected into the bilaterally hippocampus of rats to establish the AD inflammatory models. Morris water maze was used to examine the ability of learning and memory, immunohistochemical assay was used to detect the expressions of iNOS and IKKβin the rat hippocampus. Results The rats in AD group displayed longer average escape latency (AEL) , less frequency of trying to find the platform and less in original platform quadrant time (P 〈 0.05). In model group, the expressions of iNOS and IKKβwas significantly increased compare with control group( P 〈 0. 01 ). The expressions of iNOS and IKKβin Chitosan-Pbosphatidylcholine group significantly decreased compare with model group (P 〈0. 05). Conclusions Chitosan-Phosphatidylcholine may inhibit the inflammatory response in hippocampus of AD rats, and improve the leanaing and memory ability of AD rats.
