乙酰胆碱受体阻断剂对脂多糖活化THP-1细胞作用的影响

摘  要:目的观察乙酰胆碱受体阻断剂对脂多糖活化人单核细胞白血病细胞THP-1分泌细胞因子的影响,探讨乙酰胆碱通路在脓毒症中作用。方法通过细胞培养,实验细胞为THP-1细胞,分别加入不同浓度银环蛇毒素培养24h后,每孔加入100ng/ml脂多糖,同时做空白对照,CO2孵箱孵育6h,离心取上清液,检测TNF-α和IL-6,结果采用SPSS 17.0进行分析。结果 THP-1呈悬浮生长,使用不同浓度的银环蛇毒单独与THP-1细胞孵育24h后,检测上清TNF-α和IL-6分泌,比较是否加银环蛇毒两组间,P分别为0.71和0.32,结果无显著性差异。加入不同浓度银环蛇毒与THP-1细胞预孵育后,再使用LPS刺激6h,在银环蛇毒浓度为10μmol/L时,TNF-α和IL-6分泌大大增加。结论银环蛇毒单独不能明显刺激THP-1细胞分泌TNF-α和IL-6,银环蛇毒对THP-1细胞Ach受体封闭存在一个剂量效应。Objective To investigate the effects of acetylcholine receptor(AehR) blocker on THP - 1 cell activated by Iipopolysac- charide(LPS) and significance of Ach pathway in sepsis. Methods THP - I cells were cultured for 24 hours with different concentra- tions bungarotoxin at 37℃ and 5% CO2 ,then 100ng/ml LPS 100μl were dispensed into all the wells and cultured for 6 hours. TNF - α and IL - 6 in the supernatant were measured by ELISA and chemiluminescence assay, respectively. The results were expressed as x + s and performed by SPSS 17.0 Microsoft pack. Results The TNF -α and IL -6 in the supernatant had no difference between the THP - 1 cell with different concentrations bungarotoxin and control(P 〉 0.05). At the 10μmol/L bungarotoxin, the TNF-α and IL -6 in the su- pernatant of THP - 1 cell activated by 100ng/ml I,PS were 6.46 ± 1. 15ng/ml and 100.7 ± 18.59pg/ml respectively, which increased deeply. Conclusion Bungarotoxin cannot cause THP - 1 cell to secret TNF - α and I1, - 6 solely, the high concentration Ach receptor blocker should inhibit the activation of THP - 1 cell with LPS.
