片仔癀对骨肉瘤MG63细胞侵袭及迁移作用的影响

摘　　要：目的:观察片仔癀含药血清对骨肉瘤MG63细胞迁移及侵袭能力的作用,对CD44v6及基质金属蛋白酶-9(MMP-9)表达水平的影响。方法:SD大鼠片仔癀溶液连续灌胃给药4d,制备含药血清,对骨肉瘤MG63细胞进行干预,应用细胞划痕实验的方法检测细胞的迁移能力,Boyden小室迁移法和明胶黏附法检测细胞的侵袭能力,用酶联免疫检测CD44v6的表达,流式细胞仪检测MMP-9的表达。结果:片仔癀含药血清组细胞侵袭及迁移能力弱,CD44v6表达值低,MMP-9表达值低,与空白组、空白血清组相比有显著性差异(P<0.01)。结论:片仔癀降低骨肉瘤MG63细胞的侵袭及迁移能力,其机制可能与降低CD44v6及MMP-9表达有关。Objective： To investigate the effect of serum with Pien Tze Huang on the migration and invasion to the expression of CD44v6 and MMP-9 in osteosarcoma MG63 cell. Methods： The rats were mediated with Pien Tze Huang liquor by gavage for four days. The drug-serum was prepared. The serum was added to culture medium of osteosarcoma MG63 cell. The migration was tested by the use of wound healing experiments; the invasion of the cell was tested by Boyden chamber migration assay and gelatin adhesion assay; the expression of CD44v6 was detected by enzyme-linked immunosorbent assay （ELISA）; the expression of MMP-9 was observed by flow cytometry （FCM）. Results： Compared with the other two groups, its ability of migration and invasion obviously decreased, and the expression of CD44v6 and MMP-9 in Pien Tze Huang group were significantly lower. There were significant differences in comparison of the blank group and blank serum group （P〈0.01）. Conclusion： Pien Tze Huang could decrease the ability of migration and invasion of osteosarcoma MG63 cell. The underlying mechanism might be associated with down-regulation of CD44v6 and MMP-9 expression.
