摘 要:从桑天牛排泄物中分离产纤维素酶的菌株并进行鉴定,对其酶学特性以及饲料粗纤维的降解效果进行研究。采用刚果红染色法筛选出1株产纤维素酶的菌株HY3,利用形态学和分子生物学进行种属鉴定,并通过DNS法测定所产纤维素酶活力,用尼龙袋法测定其对三种饲料(麸皮、麦草秸秆、燕麦秸秆)粗纤维的降解率。结果表明,菌株HY3为地衣芽孢杆菌,此菌株所产纤维素酶的最适反应温度为70℃,最适反应pH值为6.5、菌株HY3对麸皮的粗纤维降解率最高(7.41%),对麦草秸秆粗纤维的降解率最差(4.89%)。结果提示,该菌株所产纤维素酶具有较好的热稳定性,不耐强酸碱;粗纤维的降解率随时间的变化不大。Screening and identifying the strain producing cellulase from the excretion of Mulberry longicorn and its enzymatic properties and degradation rate of the crude fiber in the feed were studied as well. A cellulase-producing bacterial strain named HY3 was screened by Con- go red staining method, identified by morphologic and biologic observation. Its enzyme activities and degradation rates of the crude fiber in three kinds of feed(bran, wheat straw, oat straw) were examined by DNS method and nylon bag method, respectively. The results showed that the strain was identified as Bacillus licheniformis, the optimum temperature of the reaction with substrate was 70 ℃, the optimum pH was 6.5, the degradation rate to crude fiber of bran was the highest(7.41%), and the degradation rate to wheat straw was the lowest(4.89%). These results indicated that the cellulase it produced had good heat stability and weak acid-base tolerance, the degradation rate of crude fiber had no significant change over time.
