摘 要:目的探讨鞘内移植骨髓基质细胞(BMSCs)对兔脊髓缺血/再灌注损伤早期水通道蛋白-4(AQP-4)表达的影响。方法日本大耳白兔72只,随机分为3组:假手术组(S组)、脊髓缺血/再灌注损伤组(I/R组)和BMSCs移植组(M组)。I/R组和M组阻断肾动脉下腹主动脉30 min建立脊髓缺血/再灌注损伤模型,S组暴露肾动脉下腹主动脉,但不阻断。损伤前2天蛛网膜下腔置管,M组注入1×108BM-SCs,I/R组注入等容量PBS。分别于再灌注损伤后4 h和24h进行神经功能评分,干湿重法测定脊髓组织含水量,测定血管外伊文思蓝含量示踪血脊髓屏障通透性变化,免疫组化和免疫印迹法检测脊髓组织中AQP-4表达情况。结果与S组比较,I/R组神经运动功能评分降低,脊髓组织含水量和伊文思蓝含量增加,AQP-4表达增加(P<0.05)。与I/R组比较,M组神经运动功能评分增高,脊髓组织含水量和伊文思蓝含量降低,血脊髓屏障AQP-4表达减少(P<0.05)。结论 BMSCs移植可抑制脊髓损伤早期AQP-4的表达,进而减轻脊髓水肿和脊髓缺血/再灌注损伤。Aim To investigate the effects of intrathecal transplantation of bone marrow stromal cells(BMSCs)on aquaporin-4 expression in spinal cord following spinal ischemia-reperfusion injury in rabbits.Methods Seventy-two Japanese white rabbits were randomly divided into 3 groups:group sham(S group);group of spinal ischemia-reperfusion injury(I/R group) and group of BMSCs(M group).Spinal ischemia-reperfusion injury was induced by clamping the abdominal aorta below renal artery for 30 min in I/R group and M group.S group was subjected to the exposure of abdominal aorta but without occlusion.I/R group and M group were intrathecally injected with PBS or BMSCs two days before injury.At 4 h and 24 h,neurological function was evaluated and damaged lumbar segment(L4-6) was removed for measurement of water content,Evans Blue content and AQP-4 protein expression.Results Compared with S group,neurological function score was significantly lower,water contents were higher and AQP-4 expression was increased in I/R group.Compared with I/R group,neurological function score was increased,water contents and Evans Blue content were lower and AQP-4 expression was decreased in M group.Conclusion Transplantation of BMSCs can inhibit AQP-4 expression after injury,and then ameliorate the spinal cord edema and spinal ischemia-reperfusion injury.
