摘 要:目的 研究白细胞介素 6 (IL 6 ) ,γ 干扰素 (IFN γ)和肿瘤坏死因子 α(TNF α)基因在甲状腺上皮细胞 (TEC)中的转录。方法 采用原代甲状腺细胞培养技术和逆转录 -聚合酶链反应 (RT-PCR)定性分析方法。结果 正常、Graves病 (GD)和多结节性甲状腺肿 (MNG)的TEC均表达IL 6mRNA ,但TNF αmRNA仅在GDTEC中表达 ,而正常和病理甲状腺细胞均未转录IFN γmRNA。结论 甲状腺细胞能够产生某些细胞因子 ,但他们在自身免疫性甲状腺疾病 (AITD)中的表达模式有异于正常甲状腺细胞 ,说明细胞因子参与AITD的发生和发展过程 ,并且对诱导AITD的免疫紊乱和甲状腺功能异常具有重要意义。Objective To clarify the different cytokine gene transcription profiles in cultured human thyroid epithelial cells. Methods Thyroid tissues were obtained from two patients with Graves' disease (GD) and two patients with multinoduler goiter (MNG). Adjecent tissues to adenomas were taken from two patients with solitary thyroid adenomas as normal controls. Thyrocytes were cultured with Eagle's medium. Reverse transcription polymerase chain reaction (RT PCR) for IL 6, IFN γ and TNF α was used. Results IL 6 mRNA was detected in both normal and abnormal thyroid cells, whereas TNF α mRNA was only transcribed in thyrocytes from GD tissues. IFN γ mRNA was not found in both normal and abnormal thyrocytes. Conclusion Thyroid cells produce many kinds of cytokines. These cytokines are important modulators for the thyroid function. The different transcription profiles in normal and pathologic thyroid tissues might contribute to the pathogenesis of autoimmune thyroid disease.
