肠三叶因子在大鼠肠道中表达的实验研究

摘　　要：目的　研究肠三叶因子 (ITF)及其mRNA在大鼠肠道的分布规律。方法　采用原位杂交、免疫组化等手段观察了ITF及ITFmRNA在大鼠肠道的表达并使用高效液相色谱仪检测了不同肠段ITF的含量。结果　从十二指肠至结肠均有ITF及ITFmRNA表达 ,主要分布于肠绒毛杯状细胞 ,其中ITFmRNA仅在杯状细胞的基底和边缘表达 ,其它区域的杯状细胞则呈ITFmRNA阴性反应 ,同时发现部分其它部位的肠上皮细胞中亦有少量ITF及ITFmRNA表达。ITF定量分析显示 ,整个肠道中以十二指肠含量最高 ,空回肠次之 ,结肠含量较低 ,各段小肠中ITF含量差异不显著 (P >0 .0 5 ) ,但都明显高于结肠 (P <0 .0 1)。结论　杯状细胞是合成ITF的主要场所 ,ITF在肠道分布的差异可能同各肠段分泌的粘液性质不同有关。Objective To study the pattern of the expression and distribution of intestinal trefoil factor (ITF) mRNA and protein in the intestines of rats. Methods The expression of ITF mRNA and protein in the intestines of rats was observed with in situ hybridization (ISH) and immunohistochemistry and the content of ITF in different regions of the intestines was measured with high performance liquid chromatography (HPLC). Results IIF and ITF mRNA distributed in the whole length of the intestines from the duodenum to the colon mainly in the base and margin of the goblet cells of intestinal villi. Goblet cells of other regions were negative of ITF expresson. Occasionaly, ITF mRNA and protein were also found in other types of intestinal epithelium. The content of ITF was 369.33±65.56, 321.66±42.12,296.45±55.23 and (186.46±30.26) ng/g in the duodenum, jejunum, ileum and colon respectively. There was no significant difference of ITF content among 3 regions of small intestine but the difference between the small intestine and the colon was statistically significant (P<0.01). Conclusion The main site to produce ITF is the goblet cells and the difference of ITF distribution in different regions of the intestines may be related to the different characteristics of the mucus secreted by the goblet cells.
