摘　　要：目的探讨Toll样受体-3（TLR3）在人肺腺癌细胞（A549细胞）和人支气管上皮细胞（HBE细胞）中的表达及意义。方法体外培养A549细胞和HB_E细胞，采用细胞免疫组织化学技术检测TLR3在2种细胞中的表达；采用实时定量聚合酶链反应（Real—timePCR）检测TLR3mRNA在2种细胞中的表达；利用Westernblot检测TLR3蛋白的表达。结果细胞免疫组织化学染色显示TLR3主要表达于两种细胞的胞膜和胞质中，呈棕黄色颗粒状，A549细胞中TLR3mRNA的表达相对值为1．01680±0．02376，高于HBE细胞，后者为0．04780±0．01792，TLR3蛋白在A549细胞中的表达相对值为0．31930±0．01116，高于HBE细胞，后者为0．16070±0．00303（P〈0．01）。结论TLR3在A549细胞中表达高于HBE细胞，提示TLR3可能参与促进人肺腺癌细胞的生长进展过程。Objective To investigate the expression and significance of Toll-like receptor 3 （TLR3） in human pulmonary adenocarcinoma cells （ A549 ） and human bronchial epithelial cells （ HBEs ）. Meth- ods The A549 cells and HBEs were cultured in vitro. The expression of TLR3 in both cells was detected by using immunoeytochemistry. The expression of TLR3 mRNA and protein in both cells was detected by using real-time quantitative polymerase chain reaction （ Real-time PCR） and Western blotting respectively. Results Immunocytochemistry staining revealed that TLR3 was mainly expressed and stained brown-yellow in cell membrane and endoehylema. The expression of TLR3 mRNA and protein in A549 cells was stronger than that in HBEs. The relative expression values of TLR-3 mRNA in A549 cells and HBE cells were 1. 0168 ±0. 02376 and 0. 0478 ±0. 01792 respectively. The relative expression values of TLR-3 protein in A549 cells and HBEs were 0. 3193 ± 0. 01116 and 0. 1607 ± 0. 00303 respectively （ P 〈 0. 01 ）. Conclusion TLR3 might cause the progression of human pulmonapy adenoeareinoma.
