雷帕霉素联合顺铂治疗人骨肉瘤的研究

摘　　要：目的比较雷帕霉素联合顺铂与单用雷帕霉素或单用顺铂对人骨肉瘤细胞增殖和凋亡的影响。方法将人骨肉瘤细胞株随机分为4组：空白组、阳性药1组、阳性药2组和治疗组。阳性药1组应用雷帕霉素20mg／L，阳性药2组应用顺铂10mg／L，治疗组联合雷帕霉素20mg/L和顺铂10mg／L，空白组加入等量生理盐水。利用噻唑蓝（MTT）比色法检测雷帕霉素联合顺铂或单用霄帕霉素川顷铂对骨肉瘤细胞的增殖抑制率；流式细胞术检测雷帕霉素联合顺铂或单川雷帕霉素／顺铂对骨肉瘤细胞凋亡率的影响。结果联合应用两种药物对人骨肉瘤的抑制牢较单独使用明显增强，且差异有统计学意义（P〈0．05），空白组：0．33±0．02，阳性药1组：0．56±0．05，阳性药2组：0．55±0．03，治疗组：0．95±0．01。单独应用雷帕霉素和顺铂，可诱导骨肉瘤细胞凋亡；联合应用时，诱导凋亡作用显著增强，且差异有统计学意义（P〈0．05）。结论雷帕霉素与顺铂联合应用，可明显抑制骨肉瘤细胞的增殖并诱导其凋亡，比单独应用效果更佳。Objective By the comparision of the rapamyein combined with cisplatin with the rapamycin or cisplatin alone towayds osteosarcoma＇ s proliferation and apoptosis, to provide new method in treating osteosarcoma in the clinical. Methods Bone sarcoma cell line were randomly divided into 4 groups： The blank group, positive medicine group 1, positive medicine group 2 and the treatment group, to add 20 mg/L rapamyein in the positive medicine group 1, to add 10 mg/L cisplatin In the positive medicine group 2, to add 20 mg/L rapamycin and 10 mg/L cisplatin in the treatment group, and to add the equal amount of normal saline in the blank group. Then detect the Inhibition rate of every group with MMT, and the apoptosis rate with flow cytometry. Results to mean ± standard deviation （x ± s） said. Results The inhibition rate and apoptosis rate of combination towards osteosarcoma enhances obviously compared to the separate application, and the difference is statistically significant（P 〈 0. 05）. The blank group： 0. 33 ±0. 02, positive medicine group 1 ： 0. 56 ±0. 05, positive medicine group 2 ： 0. 55 ±0. 03, The treatment group ： 0. 95 ±0. 01. The apoptosis rate of combination towards osteosareoma enhances obviously compared to the separate application, and the difference is statistically significant（ P 〈 0. 05 ）. Conclusion The combination of rapamycin and cisplatin can Inhibit osteosarcoma cell proliferation and induce its apoptosis obviously, better than the separate application, then can trear the osteosareoma better.
