摘　　要：目的探索建立B16细胞株家兔角膜层间接种模型,用于抗肿瘤药物疗效评价研究,并探寻观察的最佳时机。方法 10只日本大耳白兔分别在眼角膜中央区注入B16细胞混悬液20μl(5×106/ml)或生理盐水(对照),裂隙灯前节照相,观察5周,处死后取眼球,病理检查,光镜下观察肿瘤细胞及角膜组织的病理改变。结果 B16细胞接种后,5～7d角膜层间出现色素,14d色素明显,21d色素浓厚,28d色素开始扩展,而后角膜水肿会掩盖部分色素,35d由于血管的长入,停止观察。结论 B16肿瘤细胞家兔角膜层间接种模型易于对肿瘤生长的观察和测量,是肿瘤研究的合适模型之一,在进行抑制肿瘤的药物观察时,观察时间以7d～28d为最佳观察时间。Objective To establish an animal pathologic model by injection B16 cells into the rabbits corneal stroma, and observe the optimal period for using the anticarcinoma drug. Methods 10 rabbit eyes were injected B16 cells（5×10^6/ml） in the cornea stroma for 20μl and were observed by slit lamp for 5 weeks. Than the rabbits were sacrificed, the corneal were examined by the pathological section to observe the cancer cells and the alteration of corneal structure. Results The pigments initiate at 5 - 7 day after injection, and than become distinct at 14d, grew dense at 21d, finally spread to peripheral corneal at 28d. At this point, some pigments are covered by corneal edema. The observation was ended at 35d, while the neovessel come into the pigment area. Conclusion The animal model of B16 cells in rabbits corneal stroma is helpful to monitor tumor growth and is one of ideal models in tumor studies. The optimal observation period is during 7d - 28d, for observing the effect of anticarcinoma drug.
