摘 要:目的探讨丹参素与川芎嗪在心血管方面的协同效应,为其复方制剂提供科学依据。方法通过大鼠颈静脉插管滴注受试药(丹参素、川芎嗪、丹参素+川芎嗪),观察对在体大鼠平均动脉压(MAP)、心率(HR)的影响;采用lange-ndorff离体心脏灌流技术对大鼠心脏灌流给予受试药,lab-chart生物信号采集仪器记录观察各受试药对左心室收缩压(LVSP)、左心室发展压(LVDP)、心率(HR)、左心室内压最大上升速率(+dp/dtmax)、左心室内压最大下降速率(-dp/dtmax)以及冠脉流量(CF)等心功能指标的影响。结果在体实验中,丹参素组平均动脉压降低约7%,川芎嗪组降低约12%,而丹参素+川芎嗪组降低约26%,但各组均对大鼠心率影响不明显。在离体心脏灌流实验中,丹参素+川芎嗪组与空白对照组、丹参素组和川芎嗪组相比,具有较强的负性肌力作用,能明显降低LVSP、LVDP、+dp/dtmax和HR,且升高-dp/dtmax和CF,其差异具有统计学意义(P<0.05)。结论丹参素、川芎嗪联合应用对心血管的效应有协同作用。Aim To investigate the synergism on Cardiovascular system between tanshinol (TSN) and tetramethylpyrazine(TMP) from traditional Chinese medicine and provide a scientific basis for their compound preparation. Methods Through neck intravenous administration (TSN, TMP and TSN + TMP), their effects on the mean artery pressure (MAP) and the heart rate (HR) of rats were observed in vivo. Meanwhile, the cardiovascular effects of TSN, TMP and TSN + TMP were investigated using the isolated Langendorff perfused heart of rats. Myocardial hemodynamic parameters were monitored and recorded, such as left ventricular systolic pressure ( LVSP), left ventrieular developed pressure ( LVDP ) , heart rate ( HR ) , the maximum change rate of left ventricular pressure rise and fall( ± dp/dtmax ) and coronary flow(CF). Results TSN could reduce MAP by 7%, TMP by 12% and TSN + TMP by 26%. But HR was not obviously affected. In the langendorff experiment, it was found that compared with the control group, TSN group and TMP group, the TSN + TMP group had a stronger negative inotropic action which could remarkably reduce LVSP, LVDP, + dp/dtmax and HR, while increase -dp/dtmax and CF(P 〈 0. 05). Conclusion The combination of the two shows synergistic or additive effect, which 'may be related to the antagonism of calcium ion channel.
