黄芩苷联合帕拉米韦体内外抗甲型H1N1流感病毒作用

摘　　要：目的评价黄芩苷与帕拉米韦联合用药体内外抗甲型H1N1流感病毒作用。方法体外试验中,以甲型H1N1流感病毒感染MDCK细胞,黄芩苷与帕拉米韦联合用药,终点稀释法检测细胞上清液病毒滴度;体内试验中,以甲型H1N1流感病毒感染BALB/c小鼠,黄芩苷灌胃,帕拉米韦肌肉注射,两者联合给药,观察试验小鼠存活情况及体重变化。试验结果以MacSynergyⅡ软件分析两种药物体内外联合作用结果。结果细胞试验中,黄芩苷与帕拉米韦联用抗甲型H1N1流感病毒在95%置信区间内的协同值为3.2,表现为相加作用;小鼠试验中,黄芩苷与帕拉米韦联用,对提高感染流感病毒小鼠的存活率和抑制其体重下降表现为显著协同作用,协同值分别为69.0和105.2。结论黄芩苷与帕拉米韦联合抗流感作用比单独使用效果好,在临床上具有重要的应用价值。Aim To evaluate combination of baicalin and 2009 H1N1 virus in vitro and in vitro antiviral efficacies of antiviral effects of the peramivir on pandemic in vivo. Methods The baicalin combined with peramivir against influenza A/Guangzhou/03/2009 （ H1 N1 ） virus in MDCK cells were evaluated by detecting the virus yield in cell culture supernatants. In in vivo experiments, the survival rate and mean weight change of BALB/c mice infected with H1N1 virus were used as indexes to evaluate the effect of the combina- tion of intragastric baicalin and intramuscular peramivir. MacSynergy II was applied to analyze the interaction of baicalin and peramivir in vitro and in vivo. Resuits The net effect across the entire surface of the combination of baiealin and peramivir against H1NI virus in vitro was a volume of 3.2 at a 95% confidence limit, which indicated an additive effect. The treatment of baicalin plus peramivir significantly improved survival rates and inhibited weight drop of mice infected with H1NI virus, when compared with monotherapies. The synergic volumes of the combination of baicalin and peramivir based upon the mouse survival data and mean weight change were 69. 0 and 105.2, respectively. Conclusion The combination of baicalin and peramivir exhibited a higher efficacy against influenza HIN1 virus infection than used alone, which suggested a potential clinical value of the combination of the two agents.
