摘 要:目的探讨乳腺癌组织乳腺癌易感基因1(BRCA-1)蛋白表达与原癌基因人类表皮生长因子受体2(HER-2)的基因扩增关系。方法分别采用免疫组化法和荧光原位杂交法检测84例乳腺癌(导管内癌32例,浸润性导管癌36例,纤维腺瘤16例)组织中的BRCA-1蛋白表达和HER-2基因扩增情况。结果 BRCA-1蛋白阳性表达63例,阳性表达率为75.0%(63/84)。BRCA-1蛋白表达与淋巴结转移有关(P<0.05)。与不同组织类型无关(P>0.05)。HER-2基因扩增阳性40例,HER-2基因扩增阴性44例。36例有淋巴结转移患者HER-2扩增阳性率为86.1%(31/36),明显高于48例无淋巴结转移患者的27.1%(13/48)(P<0.01)。HER-2基因扩增阳性组BRCA-1蛋白阳性表达率为90.0%(36/40),高于HER-2基因扩增阴性组的22.7%(10/44)(P<0.01)。结论 BRCA-1蛋白表达、HER-2基因扩增与乳腺癌的发生发展有关,BRCA-1蛋白表达与HER-2基因扩增可能存在相关性。Objective To investigate the relationship between protein expression of breast cancer susceptibility gene 1(BRCA-1) and gene amplification of human epidermal growth factor receptor 2(HER-2) from surgical specimens of patients with breast cancer .Methods Immunohistochemistry and chromogenic in situ hybridization were used to detect protein expressions of BRCA-1 and gene amplification of HER-2 in 84 tumor tissues from breast cancer patients including 32 cases of intraductal carcinoma ,36 cases of infiltrating ductal carcinoma ,and 16 cases of fibroadenoma, respectively.Results The positive expression of BRCA-1 protein was found in 63 cases,the positive rate was 75.0%(63/84).The protein expressions of BRCA-1 in the different tumor histology type groups showed no significant difference (P〉0.05),but in different lymph node metastasis were significantly different (P〈0.05).Forty cases showed positive HER-2 gene amplification and 44 cases showed negative HER-2 gene amplification.The positive rate(86.1%,31/36) of HER-2 gene amplification in the breast cancer patients with lymph node metastasis was higher than that in the patients (27.1%,13/48) without lymph node metastasis(P〈0.01).The positive rate of BRCA-1 protein(90.0%,36/40) in the group with HER-2 gene amplification was higher than that(22.7%,10/44) in the group without HER-2 gene amplification (P〈0.01).Conclusion The protein expression of BRCA-1 and HER-2 gene amplification correlate with the development and progression of breast cancer.The protein expression of BRCA-1 may be related to HER-2 gene amplification.
