摘  要:目的观察5-氟尿嘧啶(5-FU)作用于p53突变型SGC-7901胃癌细胞系后,抑制细胞增殖及p53β,p53,Bax表达的变化,探讨其发生机制。方法不同浓度5-FU作用于SGC-7901胃癌细胞系,采用MTT法检测细胞的抑制率;巢式逆转录多聚酶链反应(nRT-PCR)检测p53β,p53,Bax在mRNA水平上表达的变化。结果MTT结果显示,5-FU对SGC-7901胃癌细胞系的增殖具有抑制作用,且呈剂量和时间双效应,不同浓度5-FU(0mg/L,25mg/L,50mg/L,100mg/L)作用于SGC-7901胃癌细胞系24,48,72h后,其抑制率逐渐增高(F=36.643,66.030,50.769,均P<0.01);RT-PCR结果显示,SGC-7901胃癌细胞系中p53β,p53,Bax在mRNA水平上的表达变化不明显(F=3.581,0.669,2.945,均P>0.05)。结论 5-FU作用于突变型p53.胃癌细胞系后,共抑制细胞增殖可能与p53突变的具体位点有关,亦可能与p53非依赖性途径的信号传递有关。Objective To study the effecls of 5-flurouracil(5-FU)on cell inhibition of proliferation amt the expression of p5313, p53 and Bax in mutant p53 SGC-7901 gastric cancer cell lines,and to investigate its mechanism. Methods SGC-7901 gastric cancer cells were interfered with different concentrations of 5-FU, and MTT assay was applied to detect the growth inhibition rates, p5313, p53 and Bax mR- NA was detected by nested reverse lrans(wiption-polymerase chain reaction(nRT-PCR).Results In MTF assay,it was shown that the SGC- 7901 gastric cancer cell lines were inhibited hy 5-FU. The growth inhibition rate was positively related to the concentration and culturing time. SGC-7901 gastric cancer cell lines were treated by different concentrations of 5-FU (0mg/L,25mg/L,50mg/L, 100mg/L) for 24,48 and 72 hours respectively, and the inhibition rate increased gradually ( F = 36. 643,66. 030,50. 769, P 〈 0.01 respectively). With RT-PCR analy- sis, the mRNA levels of f)53 β, p53 amt Bax were m)t changed significantly with the growing 5-FU concentra!ions ( F = 3.581,0. 669,2. 945, P 〉 0.05 respectively). Conclusion The inhibition effects of cell proliferation of mutant p53 gastric cancer cell lines treated by 5-FU may be associated with p53 mutation sites or its signal of the p53-independent pathway.
