食管癌中TOP2A的表达及临床意义

摘　　要：目的探讨TOP2A在食管癌和癌旁组织中的表达及其与食管癌发生发展、侵袭转移的关系。方法采用免疫组织化学(SP法)染色法及逆转录-聚合酶链反应(RT-PCR法)对62例食管癌(食管癌组)及38例相应的癌旁组织中(对照组)的TOP2A蛋白及转录水平的表达进行检测,并行统计学分析。结果 TOP2A蛋白在食管癌组织中的表达高于癌旁组织(对照组),且与癌组织的浸润程度、淋巴结转移、癌组织分化程度及临床预后密切相关(P<0.05)。TOP2A基因的mRNA及蛋白表达与肿瘤分级、淋巴结情况相关。结论 TOP2A的表达与食管癌发生发展、侵袭转移关系较密切,可作为判断食管癌患者诊断、治疗及生存预后的病理学指标之一。Objective To discuss the correlation between the expression of topoisomerase 2 alpha （TOP2A） and the biological Behaviors （cancer occurrence, development, invasion and metastasis） of human esophageal cell carcinoma （ESCC）. Methods Immunohistochemistry and reverse transcription polymerase chain reaction （RT-PCR） was used to detect the expression of TOP2A in 62 cases of ESCC specimens and 38 cases of non-cancerous tissue. The results were used for statistical analysis. Results The protein and mRNA expressions of TOP2A were significantly correlated to the invasion dept, lymphatic transmission and prognosis （P 〈0.05） and the differentiation of the tumor was also significant. The expression of TOP2A in esophageal carcinoma was higher than the cases of non-cancerous tissue which lie more than 5 centimeter on the ESCC tissue （P 〈0.05）. Conclusion TOP2A plays an important role in the invasion and metastasis of the esophageal carcinomas, and may serve as a valuable indicator for the diagnosis, treatment and the prognostic evaluation of the malignant.
