GGTase-1在人脑胶质瘤中的表达及其意义的研究

摘　　要：目的:研究GGTase-1在人脑胶质瘤中的表达,并分析其表达与胶质瘤病理级别的关系,探讨GG-Tase-1对人脑胶质瘤发生、发展所起的作用,从而为胶质瘤的治疗提供一定的理论依据。方法:分别应用半定量RT-PCR法和WesternBlot法检测15例人脑胶质瘤标本和4例正常脑组织中的GGTase-1β亚单位(GGTβ)mR-NA水平和蛋白水平;应用免疫组织化学法检测29例人脑胶质瘤组织标本和6例正常脑组织中GGTβ蛋白表达与定位情况。结果:GGTβmRNA和蛋白在各级人脑胶质瘤和正常脑组织均可表达,但在人脑胶质瘤中的表达明显高于正常脑组织(P<0.05);并且GGTβmRNA和蛋白在人脑胶质瘤Ⅲ～Ⅳ级组中的表达明显高于Ⅰ～Ⅱ级组(P<0.05)。结论:①GGTase-1在人脑胶质瘤和正常脑组织中均表达,但在人脑胶质瘤中呈明显高表达,可能与人脑胶质瘤的发生、发展有关。②GGTase-1的表达与人脑胶质瘤的病理分级相关。Objective：To exam the expression of GGTase-1 in human glioma tissues and discuss the significance of GGTase-1 in the pathogenesis of human glioma. Method： The mRNA and protein level of GGTase-1β subunit （GGTi3） were analyzed by RT-PCR and Western blot in 15 cases of glioma samples and 4 cases of normal brain tis-sues respectively. Immunohistochemistry was used to detect the protein level and location of GGTβ in 29 cases of glioma samples and 6 cases of normal brain tissues. Result：The GGTβmRNA and protein expression can be detec-ted both in normal and glioma samples. The GGTβlevels in glioma were higher than that expressed in normal brain tissues（P〈0.05）. Moreover, the mRNA and protein levels of GGTβ were gradually increased with the grade of glioma increasing （P〈0. 05）. Conclusion： GGTase-1 levels in glioma were higher than that expressed in normal brain tissues, and the expression level was positively related with the tumor grade. The abnormal expression of GG-Tase-1 may play an important role in the occurrence and develooment of glioma.
