摘  要:目的 探讨外源性透明质酸 (HA)对 C6脑胶质瘤细胞大鼠脑内移植瘤增殖及侵袭特性的影响 .方法 绿色荧光蛋白 (EGFP)基因体外转染 C6胶质瘤细胞 ,立体定向法接种入 SD大鼠脑内建立大鼠脑内移植瘤模型 ,其中实验 A,B,C组大鼠接种的细胞琼脂悬液中分别含有 5 0 ,10 0和 15 0 μg透明质酸 ,对照组接种时不给以透明质酸 . MRI定期检测移植瘤生长情况 ,并计算移植瘤生长潜伏期和体积倍增时间 . 4wk后处死大鼠作鼠脑连续石蜡切片 ,荧光显微镜检测移植瘤的对侧大脑半球和远处侵袭情况 .统计学方法比较实验组和对照组间的移植瘤生长潜伏期、体积倍增时间、对侧大脑半球侵袭率及最远侵袭距离 .结果 实验 B,C组移植瘤生长潜伏期及体积倍增时间分别为 (7.3± 2 .5 ) d,(3.8± 1.6 ) d和(6 .2± 1.6 ) d,(3.3± 1.5 ) d,明显短于对照组的 (9.1± 3.3)d,(5 .5± 2 .2 ) d(P<0 .0 5 ) ,而实验 A组与对照组间无统计学差异 (P>0 .0 5 ) ;实验 B,C组移植瘤最远侵袭距离分别为(8.9± 4.2 ) mm,(9.7± 4.7) mm,明显远于对照组 (7.1±2 .3) mm (P<0 .0 5 ) ,而实验 A组与对照组间无统计学差异(P>0 .0 5 ) ,实验组的对侧半球侵袭率和对照组无明显差异 ,均为 10 0 % .结论 一定剂量的外源性 HA对AIM To study the effects of exotic hyaluronic acid on the proliferation and invasion of transplanted glioma established by implanting C6 cells into brain of SD rats. METHODS The C6 cells which were transfected with plasmid vector containing EGFP gene were stereotacticly implanted into SD rats brain to establish the transplanted glioma model. Hyaluronic acid in different doses was mixed with cell agar suspension in groups A, B, C, while it did not exist in the control group. The growth of transplanted glioma was monitored by MRI, and its latent period and volume doubling duration were calculated. Rats were killed four weeks later and their brains were obtained to make continuous sections. The brain sections were examined using fluorescence microscopy for the visualization and detection of tumor cell invasion. The latent period, volume doubling duration, ratio of tumor cells invading into contralateral hemisphere and the longest invasion distance were compared between experimental groups and control group with statistic method. RESULTS The latent period and volume doubling duration of experimental groups B, C were (7.3±2.5) d, (3.8±1.6) d and (6.2±1.6) d, (3.3±1.5) d respectively, obviously shorter than those of control group [(9.1±3.3) d, (5.5± 2.2 ) d , P <0.05], but there was no evident difference of the latent period and volume doubling duration between experimental group A and control group( P >0.05). The longest invasion distance of B, C experimental groups were (8.9± 4.2 ) mm and (9.7±4.7) mm respectively, significantly longer than that of control group [(7.1±2.3) mm, P <0.05], while there was no evident difference of the longest invasion distance between experimental group A and control group ( P >0.05). Nor was there any evident difference in the ratio of glioma cells invading into the contralateral hemisphere between experimental and control group.
