摘 要:目的:研究30mJ/cm2的UVB照射后,HaCaT细胞周期的改变及其可能机制。方法:以人永生化上皮细胞(HaCaT)为研究对象,波长305nm的UVB为干预因子,首先观察了HaCaT细胞在30mJ/cm2的UVB作用后其细胞周期的变化;其次观察了UVB照后不同时间点p53蛋白、14-3-3σ蛋白表达的改变;最后用miR-365高表达的HaCaT细胞分析了miR-365在UVB所致了的周期阻滞中的可能作用。结果:HaCaT细胞在30mJ/cm2的UVB作用后18h出现较明显的G2/M期阻滞;p53、磷酸化的p53以及14-3-3σ蛋白在UVB照射后均有明显升高;miR-365高表达的HaCaT细胞,14-3-3σ蛋白的mRNA表达下降,且在UVB照射后无明显改变。结论:30mJ/cm2的UVB照射可诱导HaCaT细胞发生G2/M期阻滞,p53、14-3-3σ、miR-365可能在其中发挥了重要作用。Objective To study the changes of cell cycle of HaCaT cells induced by 30 mJ /cm2 UVB irradiation and its possible mechanism.Methods The cell cycle of human epithelial cells(HaCaT)was first analyzed after irradiated by 30 mJ /cm2 of UVB(305 nm);then the expressions of p53,phosphorylated p53 and 14-3-3σ protein at different time points,and the role of miR-365 in cell cycle arrest induced by UVB irradiation of miR-365 high-expressing HaCaT cells were observed.Results Obvious G2 /M phase arrest was observed in HaCaT cells at 18 h after 30 mJ /cm2 of UVB irradiation.The expressions of p53,phosphorylated p53 and 14-3-3σ protein were significantly increased after UVB irradiation.The expression of 14-3-3σ mRNA was decreased in miR-365 high-expressing HaCaT cells,and had no obvious change after UVB irradiation.Conclusion G2 /M phase arrest of HaCaT cells could be induced by 30 mJ /cm2 UVB irradiation,and p53,14-3-3σ and miR-365 may play important roles in this process.
