小鼠固化瘤苗诱发的抗肿瘤免疫实验研究

摘　　要：目的 :探讨高温固化瘤苗诱发的抗肿瘤效应 ,为其治疗肿瘤患者提供依据。方法 :用 6 5℃ / 15 m in处理 S180肿瘤细胞并制备成固化瘤苗 ,应用小鼠为实验对象 ,通过 TCR- STV (固化瘤苗 ) - MHC三分子复合体实验 ,观察其在体外诱导淋巴细胞杀伤活性 ;通过免疫预防接种和免疫治疗荷瘤实验 ,观察其在小鼠体内诱发的抗瘤效应。结果 :固化瘤苗体外诱发淋巴细胞杀伤活性为 78.8% ;用其免疫小鼠可使小鼠获得对随后接种同源细胞攻击 ,有免疫保护作用 (免疫保护率达80 % ) ,并产生免疫记忆 ;对照组小鼠未产生免疫保护作用 ,用其治疗荷瘤小鼠时 ,也明显抑制肿瘤生长 (抑制率为 86 .0 2 % ,P <0 .0 0 1)。结论 :6 5℃ / 15 m in处理 S180肿瘤细胞制成瘤苗能激活宿主免疫系统 ,抑制肿瘤生长 。Objective:Anti tumor effects induced by hyperthermal solid tumor vaccine (HSTV) were explored in order to provide the evidence for the application of HSTV to treat the patients with tumor Method:HSTV was prepared by the procedure of 65℃/15min and ultrasonic emulsification from S180 rat tumor cell Lymphocyte kill activity from bearing-S180 rat spleen cells in vitro and anti-tumor in bearing S180 rat in vivo were observated by TCR-HSTV-MHC complex test and i p or i h injection of HSTV,respectively Result:Lymphocyte kill activity induced by HSTV was 78 8% compared experimental group with control one There were immune protection effect against following i H injection of S180 tumor cells and immune memory in experiment group,not in control one;and tumor growth inhibition rate was 86 02% compared experimental group with control one Conclusion:HSTV can activate rat immune system and inhibit tumor growth in vivo It is expected that HSTV is a helpful biotherapy method
