摘  要:目的探讨γδT细胞对人宫颈癌HeLa细胞增殖和凋亡的影响。方法从宫颈癌患者手术切除的肿瘤组织分离肿瘤浸润淋巴细胞,固相抗体包被法体外扩增得到γδT细胞(效应细胞),与人宫颈癌HeLa细胞(靶细胞)按照不同的比例共培养24h或48h后,MTT法检测HeLa细胞增殖情况,流式细胞术(FCM)检测细胞的凋亡。结果γδT细胞与HeLa细胞按不同效靶比共培养24h和48h后,MTT法检测结果显示,增殖率均较对照组显著降低(P<0.05)并显示了剂量依赖性,效靶比为32∶1时,50%以上的细胞停止增殖;γδT细胞与HeLa细胞(效靶比为4∶1)共培养24h后FCM分析结果显示,HeLa细胞凋亡率较对照组显著增强(P<0.01)。结论γδT细胞对人宫颈癌HeLa细胞增殖有抑制作用并呈现剂量依赖性,对细胞凋亡有促进作用。Objective To investigate the effect of γδT cells on the proliferation and apoptosis of human cervical cancer HeLa cells. Methods γδT ceils from TILs ( tumor infiltrating lymphocytes) of the human cervi- cal cancer specimens were expanded using the solid-phase antibody coated method. Expanded γδT cells ( effec- tor cells) co-cultured with HeLa ceils (target cells) in different ratios for 24h or 48h ,then HeLa cells prolifera- tion was measured by MTr assay and apoptosis was measured by flowcytometry (FCM). Results When expand- ed γδT cells as effector cells(E) co-cultured with HeLa cells that were set as target cells(T) in different E/T ratios for 24h or 48h,the MTT results showed γδT cells markedly inhibited cell proliferation of HeLa cells in dose dependent manners when compared with the control group( P 〈0.05 ). More than 50% of HeLa cells stops proliferating at E/T ratios of 32:1 ;the results of FCM showed apoptosis rate of HeLa cells was significantly in- creased at E/T ratios of 4:1 for 24h when compared with the control group( P 〈 0.01 ). Conclusion γδT cells could inhibit the proliferation of human cervical cancer HeLa ceils in dose dependent manners and induce apop- tosis.
