摘 要:目的 研究硫酸酯化及羧甲基化虎奶多糖的抗病毒作用效果及作用机理 ,探讨虎奶多糖应用于因柯萨奇病毒 (CVB3)导致克山病疾病预防的可能性。方法 采用病变抑制法 ,在培养的 Hela细胞上测定硫酸酯化和羧甲基化虎奶多糖对 HSV- 2和 CVB32种病毒的抑制作用 ;用 EL ISA法测定这些多糖对 HBs Ag的破坏作用。结果 2种不同取代度的羧甲基化虎奶多糖及 3种不同取代度的硫酸酯化虎奶多糖在浓度分别为 0 .5、0 .2 5、0 .12 5mg/ml时均能明显抑制有囊膜病毒 HSV - 2引起的细胞病变 ,取代度对其抗病毒活性无明显影响 ;但对无囊膜病毒 CVB3无明显的抑制作用 ,对 HBs Ag无破坏作用。结论 虎奶多糖衍生物具有很好的抗 HSV- 2病毒活性 ,其作用机理可能在于破坏病毒体的囊膜。Objective To study the antivirus activity and the mechanism of carboxymethylated and sulfonated Hunai polysaccharide.Methods The inhibitory activities of the carboxymethylated and sulfonated Hunai polysaccharides against CVB_3,HSV-2 and HBsAg were evaluated in cell culture systems or with ELISA,respectively.Results The carboxymethylated and sulfonated Hunai polysaccharides (both with different degrees of substitution,DS) could inhibit HSV-2 virus in cell culture in the concentrations of 0.5,0.25 and 0.125 mg/ml.There was little difference in their inhibitory effect among the polysaccharides with different DS;but all of them exhibited no inhibition against CVB_3 or HBsAg.Conclusions All the polysaccharides exhibited strong inhibitory activity against HSV-2 in cell culture,but they have no inhibitory effect against CVB_3 in cell culture or HBsAg by ELISA.The results suggest that polysaccharides may inhibit the growth of virus by destroying their envelopes.
