摘 要:目的研究受体型酪氨酸激酶变异体RONΔ160表达对结肠癌细胞迁移、浸润能力的影响。方法将携有受体型酪氨酸激酶RON变异体RONΔ160 cDNA的质粒转染入结肠癌细胞株RKO细胞,挑选稳定转染克隆,以过河实验和Transwell趋化运动实验检测细胞迁移能力;Matrigel基质浸润实验检测细胞浸润能力,Western blot检测E-钙粘连蛋白(E-cadherin)表达的变化。结果转染RONΔ160后RKO细胞的过河时间为(38.00±1.63)h。明显短于未转染组与载体对照组的(69.50±2.52)h与(72.00±1.63)h,差异均有统计学意义(均P<0.05)。转染后RKO细胞的穿膜能力显著增强,穿膜细胞数为(59.22±6.67)个/HP、未转染组为(25.90±4.56)个/HP、载体对照组为(28.33±6.75)个/HP,差异均有统计学意义(均P<0.05)。转染RONΔ160后,RKO细胞中E-cadherin表达降低(P<0.05)。结论 RONΔ160转染可以降低E-cadherin表达,降低肿瘤细胞间的黏附性,增加结肠癌细胞RKO的移动、浸润能力。RONΔ160的表达可能是结肠癌浸润、转移的机制之一。Objective To investigate effects of receptor tyrosine kinase RON△160 on motile/invasive ability of human col-orectal cancer cel line RKO. Methods The eukaryotic expression vector pcDNA3.1-RON△160 was transfected into colorectal cancer RKO cel s and a stable expression clone was obtained. The motile ability of RKO cel s was assessed by wound healing test and the Transwel migration assay;the invasive ability was tested by Matrigel invasion method and the expression of E-cad-herin was detected by Western blot. Results Transwel chemotaxis assay revealed that the motile ability of transfected RKO cel s was greatly enhanced (P〈0.01). The wound healing time of transfected RKO cel s (38.00±1.63h) was significantly shorter than those in parental cel s (72.00±1.63h) and control cel s (69.50±2.52h)(P〈0.05). The penetrated cel s for above three groups were 59.22±6.67/HP, 25.90±4.56/HP and 28.33±6.75/HP, respectively in the Matrigel invasion test (P〈0.01). E-cadherin ex-pression decreased statistical y in pcDNA3.1-RON△160 transfected cells (P〈0.05). Conclusion Transfection of RON△160 to colorectal cancer RKO cel s can decrease of E-cadherin expression and cel-cel adhension, while migration/invasion ability is enhanced, indicating RON△160 might play roles in migration/invasion of colorectal cancer.
