摘 要:目的 研究硒 (Se)作用后淋巴细胞对实验性裸鼠大肠癌的治疗效果及其抗肿瘤作用机制。方法 建立大肠癌细胞株LoVo裸鼠移植瘤的动物模型。将荷瘤裸鼠 72只随机分成对照组、硒组、淋巴细胞组、淋巴细胞 +硒组。观察大肠癌移植瘤的生长以及荷瘤动物的肿瘤发生率和生存率 ,采用免疫组织化学法检测肿瘤组织Fas/FasL的表达情况。结果 各组肿瘤体积分别为 (1.970±0 .5 6 6 )mm3 、(1.2 90± 0 .395 )mm3 、(0 .490± 0 .2 0 8)mm3 、(0 .2 70± 0 .139)mm3 ;肿瘤发生率分别为91.6 6 %、75 .0 0 %、5 0 .0 0 %、33.33% ;生存率分别为 16 .6 6 %、33.33%、5 0 .0 0 %、75 .0 0 %。肿瘤组织免疫组织化学结果表明 ,经硒作用后肿瘤细胞Fas和淋巴细胞FasL表达增强。结论 硒有增强淋巴细胞大肠癌细胞移植瘤的作用。Objective\ To study the inhibitory effect of lymphocytes enhanced by selenium on xenotransplanted colorectal neoplasm in nude mice and the antitumor mechanism. Methods\ The animal model of xenotransplanted colorectal cancer of the LoVo cell line was established in 72 nude mice. The 72 tumor-bearing nude mice were randomly divided into the following groups: control group, Se group, lymphocyte group, lymphocyte+Se group. The growth of xenotransplanted colorectal cancer and the tumor incidence and survival rate in the tumor-bearing nude mice were observed. Immunocytochemical method was used to detect Fas/FasL expression in tumor tissue. Results\ The volume of tumor in various groups was (1.970±0.566) mm 3、(1.290±0.395) mm 3、(0.490±0.208) mm 3、(0.270±0.139) mm 3、the incidences of the tumor 91.66 %, 75.00 %, 50.00 %, 33.33 %, and the rate of survival 16.66 %、 33.33 %、 50.00 %、 75.00 %. Immunocytochemistry analysis revealed that both Fas expression in tumor cells and FasL expression in lymphocytes of tumor tissue was increased after lymphocyte+Se therapy. Conclusion\ Selenium could enhance the inhibitory effects of lymphocytes on xenotransplanted colorectal cancer.\;
