摘 要:目的 观察神经损伤后外源性表皮生长因子 (EGF)对神经元保护的情况。方法 雄性SD大鼠18只 ,体重 180~ 2 2 0 g ,随机分成对照组和EGF组 ,每组 9只。将大鼠左侧坐骨神经在梨状肌下缘 0 .5cm处切断 ,近侧端双重结扎。对照组于神经结扎近端注入生理盐水 5 μl、EGF组注入hEGF5 μl( 10 μgEGF溶于 5 μl生理盐水中 )。分别于手术后 7、14、2 8d取L4 6脊髓及L5背根神经节作苏木精 伊红染色及焦油紫染色后计算脊髓内神经元的数目 ,TUNEL标记检测 ,标记凋亡的神经元数目。结果 各时间组的神经元数目 ,EGF组明显多于对照组 (P <0 .0 1)。凋亡细胞在术后 7d、14d时 ,EGF组少于对照组 (P <0 .0 5 ) ,在 2 8d时两组差异无显著性 (P >0 .0 5 )。Objective To investigate the protective effect of exogenous Epidermal Growth Factor (EGF) on neuronal damage. Methods Eighteen male SpragueDawely rats, 180~220 g weight (8~9 weeks of age) were randomly divided into two groups, one of which as control and the other treated with EGF. Sciatic nerve injury model was set up by transection of left sciatic nerve at 0.5 cm away from pyriform muscle. The proximal stump of sciatic nerve was ligated to inhibit nerve regeneration. Then 5 μl of normal saline and an equal volume of EGF solution (containing 10 μg EGF) were injected into the proximal end of the ligated nerve in the control group and in the EGF treated group, respectively. The rats were sacrificed at 7, 14, 28 days postoperatively respectively, then perfused with 4% parafromaldehyde. The L46 spinal cord and L5 DRG were harvested, dyeing with HE, CV technique for counting total number of motor neurons and compared with those of the contralateral side. TUNEL technique was used for detection of apoptotic motor and sensory neurons. Results The number of motor neurons was significantly higher in EGFtreated group throughout the duration than that in the control group. There was significant difference between the two groups (P<0.01). The number of apoptotic motor and sensory neurons were significantly less at 7、14 d in EGFtreated groups than that in the control group (P<0.05). But there was no difference between the two groups at 28 d (P>0.05).Conclusion EGF has protective effect on the corresponding neurons after nerve injury.(Shanghai Med J, 2000,23∶411413)
